The aim of the study is to examine the effects of agmatine supplementation and exercise on cognitive functions of aged rats. In the study, 36 healthy, old (20 months) male Sprague-Dawley rats with a weight of 300-350 gr were used. The rats were randomly divided into as control (n=6), exercise (n=10), agmatine (n=10), agmatine+exercise (n=10). All the subject groups were fed with standard rat food during the studies. No application was carried out to the rats in the control group. 40 mg/kg Agmatine (i.p) supplementation was given to the rats at the agmatine groups. Swimming exercise was applied for 8 weeks to the exercise and agmatine+exercise groups. At the end of the 8th week Morris maze test was concluded. All the animals were decapitated after the test. The blood and tissue samples were taken for measuring the agmatine, brain derived neurotrophic factor and nitrite+nitrate levels.Agmatine level was significiantly higher in the agmatine and agmatine+exercise groups compared to the control group and exercise group (p ˂ 0,05). In terms of the BDNF and nitrite+nitrate levels there was no significant difference between the groups (p ˃ 0,05). On the first day, there was no difference between the groups in terms of total logged distance at the phase of Morris water maze test (p ˃ 0,05). On the second day, the total logged distance was lower in the agmatine+exercise group compared to the control group (p ˂ 0,05). On the third day, the total logged distance was lower in the groups of both agmatine and agmatine+exercise groups compared to the control group (p ˂ 0,05). On the fourth day, there was no difference between the groups in terms of total logged distance (p ˃ 0,05). In terms of finding the platform there was no significant difference between the groups (p ˃ 0,05) on the very first day. On the second day, the total logged distance was lower in the agmatine+exercise group compared to the control group (p ˂ 0,05). On the third day, the total logged distance was lower in the groups of both agmatine and agmatine+exercise groups compared to the control group (p ˂ 0,05). On the fourth day, there was no difference between the groups in terms of total logged distance (p ˃ 0,05). In terms of finding the platform there was no significant difference between the groups (p ˃ 0,05) on the very first day. On the second day there was a significant decrease at the agmatine+exercise group in terms of durations of finding the platform compared to the control group (p ˂ 0,05). On the third day there was a significant decrease in terms of platform finding durations at both the agmatine and the agmatine+exercise groups compared to the control group (p ˂ 0,05). On the fourth day there was no difference between the groups in terms of finding platform (p ˃ 0,05). There was no significant difference between the groups in total logged distance, platform reaching duration, average speed, duration passed at the platform region, platform passing count and frequency of reaching to the platform region at the Morris swimming test (p ˃ 0,05). Consequently, it was observed that agmatine supplementation might have positive effects on cognitive functions in aged rats. Key Words: Agmatine; BDNF; Exercise; Learning.

Çalışmanın amacı, yaşlı sıçanlarda agmatin takviyesi ve egzersizin kognitif fonksiyonlar üzerine etkilerinin incelenmesidir.Çalışmada, 300-350 gr ağırlığında, 36 adet sağlıklı yaşlı (20 aylık) erkek Sprague-Dawley sıçan kullanıldı. Sıçanlar rastgele olarak kontrol (n=6), egzersiz (n=10), agmatin (n=10), agmatin+egzersiz (n=10) gruplarına ayrıldı. Bütün gruplar çalışma boyunca standart sıçan yemi ile beslendi. Kontrol grubunda yer alan sıçanlara herhangi bir uygulama yapılmadı. Agmatin gruplarında bulunan sıçanlara 8 hafta 40 mg/kg agmatin (i.p) takviyesi uygulandı. Egzersiz ve agmatin+ egzersiz gruplarına 8 hafta boyunca yüzme egzersizi uygulandı. 8. haftanın sonunda Morris su labirent testi gerçekleştirildi. Testten sonra tüm hayvanlar dekapite edildi. Agmatin, Beyin kaynaklı nörotrofik faktör, nitrit+nitrat seviyelerinin ölçümü için kan ve doku örnekleri alındı. Agmatin seviyesi agmatin ve agmatin+egzersiz gruplarında kontrol grubu ve egzersiz grubuna göre belirgin şekilde yüksekti (p ˂ 0,05). BDNF seviyesi ve nitrit+nitrat seviyeleri bakımından gruplar arasında belirgin bir fark yoktu (p ˃ 0,05). Morris su labirent testi deneme fazında 1.gün gruplar arasında toplam katedilen mesafe açısından fark yoktu (p ˃ 0,05). 2.gün toplam kat edilen mesafe kontrol grubuna göre agmatin+egzersiz grubunda daha düşüktü (p ˂ 0,05). 3.gün toplam kat edilen mesafe kontrol grubuna göre hem agmatin hem de agmatin+egzersiz gruplarında belirgin olarak daha düşüktü (p ˂ 0,05). 4.gün gruplar arasında toplam kat edilen mesafe açısından fark yoktu (p ˃ 0,05). Platformu bulma süreleri açısından 1.gün gruplar arasında platformu bulma süreleri açısından anlamlı bir fark yoktu (p ˃ 0,05). 2.gün platformu bulma süreleri açısından kontrol grubuna göre agmatin+egzersiz grubunda anlamlı bir azalma vardı (p ˂ 0,05). 3.gün platformu bulma süreleri açısından kontrol grubuna göre hem agmatin hem de agmatin+egzersiz grubunda anlamlı bir azalma vardı (p ˂ 0,05). 4. gün gruplar arasında platformu bulma süreleri açısından fark yoktu (p ˃ 0,05). Morris yüzme testinin öğrenme fazında toplam kat edilen mesafe, platforma ulaşma süresi, ortalama hız, platform bölgesinde geçirilen süre, platform üzerinden geçme sayısı ve platform bölgesine uğrama sıklığı açısından gruplar arasında anlamlı fark yoktu (p ˃ 0,05). Sonuçta, agmatin takviyesinin yaşlı sıçanlarda kognitif fonksiyonlar üzerine olumlu etkileri olabileceği gözlemlendi.Anahtar Sözcükler: Agmatin; BDNF; Egzersiz; Öğrenme.
