Boric acid, one of the most common bor compounds, has antioxidant and anti-inflammatory properties, and is widely used as an additive in medicinal products. The present study was carried out to investigate the protective effects of boric acid in the ischemia/reperfusion (I/R)-induced acute kidney injury model and to determine its effect on oxidative damage, inflammatory, and apoptosis. To evaluate the protective effects of boric acid, renal I/R model was induced in rats by 50 minutes of ischemia followed by 3 h of reperfusion. Six groups of rats were studied: I; control (laparotomy was performed), II; renal I/R, III: single dose 7 mg/kg boric acid, IV; single dose 14 mg/kg boric acid, V; 7 mg/kg boric acid-treated 1 hour before induction of renal I/R, VI; 14 mg/kg boric acid-treated 1 hour before induction of renal I/R. The oxidant/antioxidant properties were evaluated using the MDA (malondialdehyde), CAT (catalase), SOD (superoxide dismutase), and GSH (glutathione). To determined the anti-inflammatory, the TNF-α (tumor necrosis factor-α) levels were performed. Kidney damages and apoptosis were also evaluated immunohistochemically by Bax and 8-OHdG (8-hydroxylo-2'-deoxyguanosin), and routine histological staining methods. Oral pretreatment of boric acid (14 mg/kg) ameliorated the renal damage compared to the control group, the reducing of cell apoptosis and histopathological changes. Moreover, boric acid (14 mg/kg) reduced inflammatory parameters and oxidative stress on renal injury. It is also important to highlight that oral treatment of boric acid (7 and 14 mg/kg) did not produce any sign of acute toxicity in rat kidneys. The data show that boric acid (14 mg/kg) has evident protective potential against renal injury, mainly through antioxidant defense, anti-inflammatory and antiapoptotic activity.

En yaygın bor bileşiklerinden biri olan borik asit, antioksidan ve antienflamatuar özelliklere sahiptir ve tıbbi ürünlerde bir katkı maddesi olarak yaygın şekilde kullanılmaktadır. Bu çalışma, borik asidin iskemi/reperfüzyon (İ/R)-indüklü akut böbrek hasarı modelindeki koruyucu etkilerini araştırmak ve bunun oksidatif hasar, enflamatuar ve apoptoz üzerindeki etkisini belirlemek için yapılmıştır. Borik asidin koruyucu etkilerini değerlendirmek için, böbrek İ/R modeli sıçanlarda 50 dakikalık iskemi ve ardından 3 saat reperfüzyon ile indüklenmiştir. Altı sıçan grubu çalışılmıştır: I; kontrol (laparotomi yapılan), II; böbrek İ/R, III; tek doz 7 mg/kg borik asit, IV; tek doz 14 mg/kg borik asit, V; renal İ/R'nun indüklenmesinden 1 saat önce 7 mg/kg borik asit uygulanan, VI: renal İ/R'nun indüklenmesinden 1 saat önce 14 mg/kg borik asit uygulanan. Oksidan/antioksidan özellikler, MDA (malondialdehit), KAT (katalaz), SOD (süperoksit dismutaz) ve GSH (glutatyon) kullanılarak değerlendirilmiştir. Antiinflamatuarın saptanması için ise TNF-α (tümör nekroz faktörü-α) düzeyleri gerçekleştirilmiştir. Böbrek hasarları ve apoptosis ayrıca immünohistokimyasal olarak Bax ve 8-OHdG (8-hidroksil-2'-deoksiguanosin) ve rutin histolojik boyama metodlarıyla değerlendirilmiştir. Borik asidin (14 mg/kg) ağız yoluyla ön uygulaması, böbrek hasarını kontrol grubuna kıyasla hücre apoptozunu ve histopatolojik değişiklikleri azaltarak hafifletmiştir. Ayrıca, borik asit (14 mg/kg), böbrek hasarı üzerindeki enflamatuar ve oksidatif stres parametreleri azaltmıştır. Borik asidin (7 ve 14 mg/kg) oral olarak uygulanmasının, sıçan böbreklerinde herhangi bir akut toksisite belirtisi üretmediğini de vurgulamak önemlidir. Veriler, borik asidin (14 mg/kg), özellikle antioksidan savunma, anti-enflamatuar ve antiapoptotik aktivite yoluyla, böbrek hasarına karşı belirgin bir koruyucu potansiyele sahip olduğunu göstermektedir.
