This study was carried out to determine the effects on the growth of cotton plants and the in-vivo and in-vitro efficacy of these bacteria against wilt disease caused by the fungus called Verticillium dahliae Kleb, which causes significant yield and quality losses in cotton, by isolating plant growth promoting rhizobacterial (PGPR) from soil samples taken from cotton production areas in Kahramanmaras province and towns with Nurdağı town of Gaziantep in 2013 (July-September). A total of 681 bacterial isolates were obtained from the soil taken from cotton plant root rhizosphere as a result of isolation studies.In in vitro experiments 243 of the total 681 bacterial isolates were determined to be phosphorus (P) solubising plant growth promoting rhizobacterial (PGPR). In addition, of the obtained isolates were determined to have antogonistic effects on the mycelial growth of V.dahliae with dual culture test method in vitro conditions. Among the tested bacterial isolates, thirty isolates were determined to inhibit mycelial growth of Vd between 3,5-55,7%. Among the all bacterial, 101 PGPR isolates, having the highest P-solubising potential and 30 the most effective antagonist isolates were selected to use in vivo pot experiments. 15 PGPR isolates showing the best effect on plant growth and 3 PGPR isolates showing the best antagonistic effect as a result of pot experiments were identified as Bacillus megaterium (1), Pseudomonas corrugata (1), Acinetobacter lwoffii (1), Enterobacter ludwigii (6), Enterobacter cloacae (2), Acinetobacter pittii (3), Arthrobacter oxydans (1), Bacillus cereus (1), and Bacillus mojavensis (2) by using biochemical tests and MALDI-TOF MS technique.Seed treatment with selected isolates were found to suppress disease severity in varying ratios of 16.14-90.04 % in the '' Beren '' cotton variety, which is susceptible against the disease and 1.4-92.2 % in the '' Teks '' cotton variety, which is tolerant against the disease, in the pot experiments under climate room conditions comparable to the only Vd experiments.In the absence of the pathogen, these isolates were also found to enhance plant growth parameter (such as plant height of cotton, number of cotton node, number of cotton total leaf, root lenght of cotton, cotton dry and wet weight) in varying ratios in pot experiments.
Bu çalışma, 2013 (Temmuz -Eylül) yılında Kahramanmaraş il ve ilçeleri ile Gaziantep ilinin Nurdağı ilçesinde pamuk üretim alanlarından alınan toprak örneklerinden bitki gelişimini teşvik eden kök bakterilerini izole etmek ve bu bakterilerin pamukta önemli verim ve kalite kayıplarına sebep olan Verticillium dahliae Kleb. adlı fungusun neden olduğu solgunluk hastalığına karşı in-vivo ve in-vitro etkinliklerini ve ayrıca pamuk bitkisi gelişimine olan etkilerini belirlemek amacıyla yürütülmüştür. Yapılan izolasyon çalışmaları sonucunda pamuk bitkisi kök bölgesinden alınan topraklardan toplam 681 adet bakteri izolatı elde edilmiştir. Toplam 681 bakteri izolatından 243 tanesinin in vitro denemelerde fosfor (P) çözücü bitki gelişimini teşvik eden kök bakterisi (PGPR) olduğu belirlenmiştir. Ayrıca, elde edilen izolatların, in vitro koşullarda ikili kültür test yöntemi ile V.dahliae'nın (Vd) misel gelişimi üzerindeki antagonistik etkileri belirlenmiştir. Test edilen bakteri izolatları arasından, 30 izolatın Vd'nin misel gelişimini % 3,5-55,7 arasında baskıladığı tespit edilmiştir. Tüm bakteriler arasından en yüksek P çözme potansiyeline sahip olan 101 adet PGPR izolatı ve en yüksek antagonistik özelliğe sahip 30 izolat seçilerek in vivo denemelerde kullanılmıştır. Saksı denemeleri sonucunda bitki gelişiminde en iyi etkiyi gösteren 15 ve en iyi antagonistik etkiyi gösteren 3 PGPR izolatı, MALDI-TOF MS tekniği ve biyokimyasal testler ile Bacillus megaterium (1), Pseudomonas corrugata (1), Acinetobacter lwoffii (1), Enterobacter ludwigii (6), Enterobacter cloacae (2), Acinetobacter pittii (3), Arthrobacter oxydans (1), Bacillus cereus (1), ve Bacillus mojavensis (2) olarak tanımlanmıştır. Seçilen izolatların tohumla inokulasyon yapılması ile sadece Vd inokulasyonu yapılmış denemeler ile karşılaştırıldığında iklim odası koşullarındaki saksı denemelerinde hastalığa duyarlı olan ''Beren'' pamuk çeşidinde hastalık şiddetini %16,14 - 90,04 ve hastalığa karşı tolerant olan ''Teks'' pamuk çeşidinde ise hastalık şiddetini %1,4-92,2 arasında değişen oranlarda baskıladığı saptanmıştır. Sera koşullarında kurulan saksı denemesinde patojenin yokluğunda, bu izolatların bitki büyüme parametrelerini (pamuk bitki boyu, pamuk boğum sayısı, toplam yaprak sayısı, pamuk kök uzunluğu, pamuk yaş ve kuru ağırlık) farklı oranlarda arttırdığı tespit edilmiştir.
