This thesis study is aimed at investigating the genetic characterization and to determine the genetic relationships between populations in the Karayaka sheep breed, one of the farm animal genetic resources of Turkey, using nine microsatellite markers. Blood samples collected from unrelated 206 head of Karayaka sheep in its breeding tract raised in Samsun, Ordu, Giresun, and Tokat provinces in the Black Sea Region of Turkey were used as study material. The genomic DNA was isolated by using an extraction kit. Microsatellite loci were amplified by Polymerase Chain Reaction (PCR) method, and then a total of 576 Post-PCR products to band quality were chosen from among agarose gel results, and selected samples were genotyped by a DNA fragment analysis instrument. The average number of alleles (Na), allelic richness (Ar), observed heterozygosity (Ho), expected heterozygosity (He), polymorphism information content (PIC), and inbreeding coefficient (FIS) for all populations was estimated respectively as Na=16,44, Ar=9,887, Ho=0,301, He=0,817, PIC=0,866, and FIS=0,633. The observed and expected heterozygosity ranged from 0,171 (Giresun) to 0.376 (Ordu) and 0,775 (Tokat) to 0,845 (Ordu), respectively. It was determined that 10,5% of the total genetic variation (FIT=66,7%) in Karayaka sheep corresponded to genetic differences among populations (FST) (P<0,05), whereas the genetic difference explained 63.0% among individuals (FIS). The FIS values that were calculated at population level ranged from 0,568 (Samsun) to 0,791 (Giresun), and they were all significant (P<0,001). For this reason, populations were not in the Hardy-Weinberg equilibrium. According to Factorial Corresponding Analysis (FCA), all populations were separated from each other, whereas Samsun population was clustered in the different place than other populations. The results obtained from this study showed that Karayaka sheep population had high-level genetic diversity and inbreeding coefficient.
Bu tez çalışmasında, Türkiye yerli çiftlik hayvan gen kaynaklarından biri olan Karayaka koyun ırkının genetik karakterizasyonunun yapılması ve koyun populasyonları arasındaki genetik ilişkilerin 9 adet mikrosatellit markör kullanılarak ortaya konması amaçlanmıştır. Türkiye Karadeniz bölgesi kıyı şeridi illerinden; Samsun, Ordu, Giresun ve Tokat il ve ilçe köylerinden Karayaka koyun ırkına ait 206 baş koyundan alınan kan örnekleri çalışma materyali olarak kullanılmıştır. Genomik DNA, ticari kit kullanılarak elde edilmiştir. Mikrosatellit lokuslar Polimeraz Zincir Reaksiyonu (PCR) metoduyla çoğaltıldıktan sonra lokus başına 16 ve her populasyon için 144 adet örnek seçilerek toplam 576 adet PCR sonrası ürün DNA fragment analiz cihazında genotiplendirilmiştir. Karayaka koyun ırkında lokus başına ortalama gözlemlenen allel sayısı (Na), allelik zenginlik (Ar), gözlenen heterozigotluk (Ho), beklenen heterozigotluk (He), polimorfik bilgi içeriği (PBİ) ve akrabalı yetiştirme katsayısı (FIS) değerleri sırasıyla Na=16.44, Ar=9.887, Ho=0.301, He=0.817, PBİ=0.866 ve FIS=0.633 olarak belirlenmiştir. Populasyon düzeyinde gözlenen (Ho) ve beklenen heterozigotluk (He) değerleri sırasıyla 0.171 (Giresun) ile 0.376 (Ordu) ve 0.775 (Tokat) ile 0.845 (Ordu) aralığında değişmiştir. Karayaka koyununda tespit edilen %66.9'luk genetik varyasyonun (FIT) %63.0'nün populasyonlar içindeki genetik farklılıktan (FIS) %10.5'inin ise populasyonlar arasındaki genetik farklılıktan (FST) (P<0.05) kaynaklandığı belirlenmiştir. Populasyon düzeyinde hesaplanan FIS değerlerinin 0.568 (Samsun) ile 0.791 (Giresun) arasında değişmek üzere istatistiksel olarak önemli (P<0.001) olduğu, bu nedenle populasyonların Hardy-Weinberg genetik dengesinde olmadığı sonucu elde edilmiştir. Faktöriyel uygunluk analizi (FCA) sonucuna göre, populasyonların birbirlerinden farklı kümelendiği Samsun populasyonunun ise diğer populasyonlardan daha farklı bir yerde kümelendiği belirlenmiştir. Çalışma sonucunda, Karayaka koyun ırkında akrabalı yetiştirme düzeyinin ve genetik çeşitliliğin yüksek düzeyde olduğu tespit edilmiştir.
