Natural venoms have always fascinated naturalists and researchers, and the most investigated one among them is snake venoms. Due to the great interest on peptide based drug development of late years, researches on animal venoms have significantly increased.The aim of this thesis is to characterize the venom components of three medicinally important Turkish vipers by mass spectrometry-based modern proteomic methods and determine the variations, to assess their cytotoxicity and to evaluate their biotechnological usage potential with a special attention on their effects on coagulation cascade and platelet aggregation.For this purpose, vipers were collected in the nature by field studies, their venoms were profiled by bioanalytical methods such as 2D-PAGE, gel filtration and reverse phase chromatography and high resolution mass spectrometry. Venom proteomes of three species included in the thesis were characterized and the protein families were identified by mass spectrometry based de novo amino acid sequencing approach.Effects of crude venoms on blood coagulation and platelet aggregation, as well as their cytotoxic activity were screened, which will guide the further studies on the bioactivity guided characterization of the venom samples. Detection of the significant anti-platelet activity of the venom of Macrovipera lebetina and Montivipera xanthina shows that these venoms are potential sources of novel bioactive molecules to be discovered and patented. Additionally, the results of the proteomic characterization studies correlated with the biological activities.This thesis is a pioneering study in toxinology field in Turkey and has created an important knowledge for the development of this field. In light of the results obtained in this thesis, purification and detailed characterization of the active peptide and protein components will be possible with further studies.

Doğal zehirler eski tarihlerden beri doğa bilimcilerin ve araştırmacıların ilgisini çekmiştir ve bunlar arasında en fazla araştırılan ise yılan zehirleri olmuştur. Son yıllarda peptit tabanlı ilaç geliştirme konusuna olan ilgi nedeniyle hayvan zehirleriyle ilgili araştırmaların sayısı artmıştır.Bu tez çalışmasının amacı, Türkiye'de yayılış gösteren tıbbi öneme sahip üç engerek türünün (Macrovipera lebetina, Montivipera xanthina, Vipera ammodytes) zehirlerini kütle spektrometresi tabanlı modern proteomik yöntemlerle karakterize ederek farklılıkları tespit etmek, sitotoksisitelerini ve koagülasyon üzerindeki etkilerini belirlemek ve biyoteknolojik kullanım potansiyellerini irdelemektir.Bu amaçla tez kapsamında yapılan arazi çalışmalarıyla türler doğadan elde edilmiş ve bu üç türün zehirleri 2D-PAGE, jel filtrasyon ve ters faz kromatografisi ve yüksek çözünürlüklü kütle spektrometresi gibi biyoanalitik teknikler ile profillenerek varyasyonları belirlenmiştir. Kütle spektrometresi kullanılarak gerçekleştirilen de novo amino asit dizileme tabanlı yaklaşım ile üç türün zehirlerinin proteomik karakterizasyonları yüksek doğrulukta gerçekleştirilerek içerdikleri protein aileleri belirlenmiştir.Zehirlerin sitotoksisiteleri, koagülasyon ve platelet kümeleşmesi üzerindeki etkileri belirlenerek, ileride yapılacak biyoaktivite rehberli karakterizasyon çalışmaları için rehber niteliğinde sonuçlar elde edilmiştir. Macrovipera lebetina ve Montivipera xanthina zehrinde belirgin olarak görülen anti-platelet aktiviteler, bu türlerin zehirlerinden patentli ürünlere dönüşecek yeni moleküllerin keşfedilme potansiyeli olduğunu göstermiştir. Ayrıca proteomik karakterizasyon sonuçları da elde edilen biyolojik aktiviteler ile uyumlu bulunmuştur.Tez kapsamında yapılan çalışmalar Türkiye'de ilk olma niteliğindedir ve bu alanın Türkiye'de gelişmesi için önemli bir bilgi birikimi oluşturmuştur. Tez kapsamında elde edilen bilgiler ışığında gerçekleştirilecek devam projeleri ile aktif peptit ve proteinlerin saflaştırılması ve detaylı karakterizasyonları mümkün olabilecektir.
