In this thesis impedimetric immunosensors were developed for the detection of Immunoglobulin G (IgG) in blood and cerebrospinal fluid (CSF) whose levels increase during autoimmune diseases. Protein A (PrA a protein with a high affinity to IgG, was immobilized onto screen-printed electrodes using gelatin, chitosan natural polymers and combinations of them due to their high biocompatibility, low biofouling and non-toxicity. Gold nanoparticles (AuNPs) were also used to increase sensitivity and decrease detection limits by means increasing the surface area. The amount of polymer and nanoparticles, crosslinker concentrations and PrA concentrations were optimized using Electrochemical Impedance Spectroscopy (EIS) and Cyclic Voltammetry (CV). Surface characterization was carried out with Scanning Electrone Microscopy (SEM) and Atomic Force Microscopy (AFM). Thermogravimetric Analysis (TGA) was used to evaluate the thermal stability of polymer and composite structure, and infrared spectroscopy (FT-IR) was used to explain the molecular structure. Reproducibility and shelf-time, which are important for practical use of immunosensors, has been evaluated. Immunosensor trials were perdformed for different antigens for the non-specific binding. Results showed PrA to be immobilized on the carrier systems efficiently. The designed immunosensors have low detection limits and show lineer response for broad ranges of IgG concentration. It has been shown that the immunosensor prepared using gelatin/chitosan-AuNPs-PrA has a lower detection limit. The developed immunosensor was used for the detection of IgG in Cerebrospinal Fluid (CSF) and serum. The immunosensor was tested in real samples with serum obtained from healty patients and those diagnosed with Multiple Sclerosis.

Bu tez çalışmasında otoimmün hastalıklar sırasında kan ve beyin-omurilik sıvısında (BOS) miktarı artan İmmünoglobülin G (IgG) tayinine yönelik impedimetrik immünosensörler geliştirilmiştir. Biyouyumlulukları yüksek, biyokirlenmeleri az ve toksik özellik göstermemeleri nedeniyle jelatin, kitosan doğal polimerleri ve bunların kombinasyonları kullanılarak hazırlanan platin perde-baskı (screen-printed) elektrotlara; IgG'ye yüksek affinite gösteren Protein A (PrA) immobilize edilmiştir. Ayrıca yüksek yüzey alanları sayesinde immünosensörlerin duyarlılığını arttırmak ve tayin limitini düşürmek amacıyla altın nanopartikülleri (AuNPs) kullanılmıştır. Polimer ve nanopartikül miktarları, çapraz bağlayıcı konsantrasyonları ve PrA konsantrasyonları Elektrokimyasal İmpedans Spektroskopisi (EIS) ve Dönüşümlü Voltametri (CV) kullanılarak optimize edilmiş; Taramalı Elektron Mikroskobu (SEM) ve Atomik Kuvvet Mikroskobu (AFM) kullanılarak yüzey karakterizasyonları gerçekleştirilmiştir. Kullanılan polimer ve kompozit yapının termal kararlılıklarını değerlendirmek amacıyla Termogravimetrik Analiz (TGA), moleküler yapılarını aydınlatmak amacıyla infrared spektroskopisi (FT-IR) yönteminden faydalanılmıştır. İmmünosensörlerin pratikte kullanılmasında önem arz eden tekrar üretilebilirlik ve raf ömrü çalışmaları yapılmış; ayrıca farklı antijenlerle immünosensör denenerek spesifik olmayan bağlanmaların etkisi araştırılmıştır. EIS, CV, SEM ve AFM ile yapılan karakterizasyon çalışmaları; PrA'nın taşıyıcı sistemlere etkin bir şekilde immobilize olduğunu göstermiştir. Tasarlanan immünosensörlerden en düşük tayin limitine sahip olan jelatin/kitosan-AuNPs-PrA immünosensörüdür. Geliştirilen immünosensör Multipl Skleroz teşhisi konulmuş hastadan ve sağlıklı kişiden alınan BOS ve serum ile test edilmiştir.
