The study investigating the effect of MarT regulator protein on the production of S. Typhimurium biofilm in in-vitro culture conditions, at the two consecutive concentration of arabinose (% 0.02 ve % 0.01) in which the production of MarT protein is promoting in the highest level, the biofilm formation showed a statistically significant decrease compared with the wild type strain. According to the result of real time PCR experiments, gene of marT was identified as the positive regulator of the marT, slsA, misL, rmbA, fidL and wzzB genes; as the negative regulator of the fliA, fliC, dps, fimA, cheA and cheM genes. When compared the biofilm formation capacity to the wild type in in-vitro conditions, the mutants, contain a mutation in the genes investigated in terms of marT regulation, showed statistically significant reductions were identified as fliA (% 59), fliC (% 40), fimA (% 46), fidL (% 25) and misL (% 25) mutants. Although the biofilm structures of all strains used in this study were detected as rdar morfotype in agar plates, inspections carried out under steromicroscope showed that the biofilms in rdar morphotype formed by mutant strains have different phenotypic features from each other and as well as from wild type strain. Among the genes examined; fimA, slsA, fliA, dps, cheA and wzzB genes were found to be having a direct effect on the pellicle formation of S. Typhimurium. Finally; the biofilm formation features of the mutant strain, in which marT expression is induced, on glass, stainless steel and polystyrene were also examined and determined that the ability to produce biofilm was decreased on all surfaces.

MarT regülatör proteininin in-vitro kültür koşullarında S. Typhimurium'da biyofilm üretimi üzerine etkisinin araştırıldığı çalışmada; MarT protein üretiminin en yüksek düzeyde teşvik edildiği ardışık iki arabinoz konsantrasyonunda (% 0.02 ve % 0.01) biyofilm üretiminin, doğal suşa oranla istatistiki açıdan anlamlı bir şekilde düştüğü saptandı. Gerçek zamanlı PZR denemeleri sonucu, marT geninin S. Typhimurium'da; marT, slsA, misL, rmbA, fidL ve wzzB genlerinin pozitif; fliA, fliC, dps, fimA, cheA ve cheM genlerinin ise negatif regülatörü olduğu belirlendi. marT regülasyonu yönünden araştırılan genler bakımından mutant suşların in-vitro koşullarda biyofilm üretme kapasiteleri doğal suş ile kıyaslandığında, istatistiki açıdan anlamlı düşmeler gösteren mutantlar; fliA (% 59), fliC (% 40), fimA (% 46), fidL (% 25) ve misL (% 25) olarak belirlendi. Bu çalışmada kullanılan tüm suşların biyofilm yapıları agar plaklarda rdar morfotipte saptanmış olmasına rağmen, steromikroskop altında yapılan incelemelerde mutant suşların oluşturdukları rdar morfotipteki biyofilmlerin, hem birbirinden ve hem de doğal suştan farklı fenotipik özellikler gösterdiği belirlendi. Pelikül oluşumları değerlendirildiğinde; incelenen genlerden fimA, slsA, fliA, dps, cheA ve wzzB genlerinin S. Typhimurium'da pelikül oluşumu üzerinde doğrudan etkili olduğu saptandı. Son olarak; marT ifadesi indüklenen mutantın cam, çelik ve polistiren üzerinde biyofilm oluşturma özellikleri incelendi ve tüm yüzeylerde biyofilm üretme yeteneğinin düştüğü belirlendi.
