Clavibacter michiganensis subsp. michiganensis is one of the most damaging pathogens in tomato production. The pathogen has been subjected to a first-degree quarantine organism in Turkey. There are great management efforts to control the pathogen but there is no resistant commercial varieties developed yet. In this study, we studied M3-9 tomato mutant plants which resistant to the most virulent isolate of Clavibacter michiganensis subsp. michiganensis Cmm2 using PCR-based mapping analysis to locate the resistant locus on M3-9 chromosomes. Therefore, M3-9 resistant tomato mutant plants were crossed with commercial susceptible Suvari variety and M3-9 × Suvari crosses were established. Then whole F1 plants were selfed them with allowing pollination from others. On 80 F2 tomato plants were inoculated in pathogenicity tests and they were analysed in the PCRs. Pathogenicity tests revealed that phenotypically 57 tomato plants were susceptible and 23 plants were resistant to Cmm2 in the F2 population. From the F2 population, chosen 9 most resistant and susceptible plants were analysed with more than 120 molecular markers to map resistant locus in the PCRs. Mapping study results have identified the most linked marker in genotypes where the marker was exactly matched with the phenotypes. The M3-9 mutant resistant locus was mapped with an INDEL marker on its 5th chromosome.

Clavibacter michiganensis subsp. michiganensis domates üretiminde karşılaşılan en zarar verici hastalık etmenlerindendir. Ülkemizde hastalık etmeni birinci derece karantinaya tabidir. Hastalık etmeni ile mücadele konusunda büyük çalışmalar yapılsa da ticari olarak dayanıklı bir çeşit geliştirilememiştir. Bu çalışmada Clavibacter michiganensis subsp. michiganensis'nin en virulent ırkı olan Cmm2 izoalatına dayanıklı M3-9 mutant domates bitkisinde PCR temelli haritalama çalışması yapılarak dayanıklılık lokusunun hangi kromozomda olduğu araştırılmıştır. Bunun için ilk olarak dayanıklı M3-9 mutant domates bitkisi Süvari ticari bitkisi ile melezlenerek M3-9 x Süvari F1 bitkisi oluşturulmuştur. Oluşturulan F1 bitkisinin kendilenmesine izin verilerek M3-9 x Süvari F2 popülasyonu domates bitkileri oluşturulmuştur. F2 popülasyonunda 80 bitkide patojenisite testi yapılmış ve patojen PCR tekniği ile tanılanmıştır. Patojenisite testi sonucu 23 bitki dayanıklı ve 57 bitki ise hassas olarak fenotipte belirlenmiştir. Patojenisite testi sonucu belirlenen en dayanıklı ve en hassas 9'ar bitkide 120'nin üzerinde markör ile haritalama çalışması yapılmıştır. Yapılan haritalama çalışması sonucu fenotipteki dayanıklılık ve hassaslık durumunu genotipte en belirgin olarak veren markör belirlenmiştir. Çalışma sonucu mutant M3-9 bitkisinde dayanıklılığın 5. kromozomda kontrol edildiği bir INDEL markörü ile belirlenmiştir.
