Bacterial cancer and bacterial wilting disease caused by Clavibacter michiganensis subsp. michiganensis in is able to result 100% yield losses particularly on tomato and other Solanaceae plants. For management of such bacterial pathogen, cultural and chemical control methods have been applied but these control methods are inadequate to the bacterial canker disease. The most efficient and environmentally friendly method to control the bacterial canker disease is to use resistant tomato varieties; however, there is no resistant tomato variety governs resistance to the bacterial disease not only in Turkey but also in the world. In this study, we intend to map previously obtained resistant M3-15 mutant with a molecular marker where its location knows on tomato chromosomes. M3-15 tomato plant were crossed with susceptible commercial tomato varieties. From these crosses F1 and F2 populations were formed and all the F1 and F2 plants are inoculated with a virulent Clavibacter michiganensis subsp. michiganensis pathogen. Pathogenicity test revealed that all tested F1 plants were susceptible whereas ¾ of F2 plants were susceptible and ¼ of F2 plants were resistant to the Clavibacter michiganensis subsp. michiganensis pathogen. From the F2 population, phenotypically the most susceptible and resistant tomato plants have chosen and PCR amplifications have conducted to map resistance locus with molecular markes with their susceptible and resistant parents. In the mapping studies, more than 100 SSR and INDEL markers were tested to match plants' phenotypes with their genotypes. Resistant locus of M3-15 resistant plant was matched with a molecular marker on chromosome 5. Hence, the resistant locus was mapped in tomato genome with a knowm molecular marker.

Bakteriyel kanser ve solgunluk hastalığı (Clavibacter michiganensis subsp. michiganensis) Solanacea familyasında özellikle domateste %100'e varabilen kayıplara neden olmaktadır. Hastalığın kontrolünde kültürel ve kimyasal mücadele yöntemleri uygulanmakta fakat etkin bir mücadele yapılamamaktadır. En etkin ve çevreci yöntem olarak dayanıklı çeşit kullanılması gerektiği bilinsede Dünya'da bu hastalığa karşı dayanıklı domates çeşidi olmadığı bilinmektedir. Bu çalışma ile daha önceden kimyasal mutasyon ile elde edilen dayanıklı M3-15 bitkisinde dayanıklılığı sağlayan lokusun kromozomlar üzerinde yerleri bilinen markörlerle haritalanması amaçlanmıştır. M3-15 domates bitkisi hassas ticari domates çeşitleri ile melezlenerek F1 ve F2 popülâsyonları oluşturulmuştur. Bu popülâsyonların Clavibacter michiganensis subsp. michiganensis ile patojenisite testlerinde F1 bitkilerinin tümü hassas, F2 popülasyonunda ise ¾ oranında hassas ve ¼ oranında dayanıklı domates bitkileri bulunmuştur. F2 popülâsyonundaki fenotipik olarak en hassas ve en dayanıklı 10'ar domates bitkileri ebeveynleriyle birlikte haritalama çalışmalarında kullanılmıştır. Haritalama çalışmalarında 100'den fazla SSR ve INDEL markırı test edilmiş olup M3-15 domates bitkisinde dayanıklılık lokusu 5 no'lu kromozom üzerindeki markırla eşleşmiştir. Böylece M3-15 domates bitkisinde Clavibacter michiganensis subsp. michiganensis'e karşı dayanıklılık lokusu domates kromozomu üzerinde haritalanmıştır.
