Atomoxetine is a selective noradrenaline reuptake inhibitor drug that is indicated for the treatment of attention deficit hyperactivity disorder. Regarding the fact that the drugs increasing the monoaminergic concentration in the brain have therapeutic potential for neuropathic pain treatment, it is planned to investigate the possible efficacy of atomoxetine on diabetes induced neuropathic pain in this study. Data from the Randall-Selitto and the Hargreaves tests indicate that diabetes caused a significant decrease in paw withdrawal threshold and paw withdrawal latency in the rats. On the other hand, atomoxetine administered at 3 mg/kg/day for 7 and 14 days, improved the aforementioned hyperalgesic responses induced by diabetes. Mechanistic studies were also carried out using α-methyl-para-tyrosine methyl ester (AMPT), fentolamine and propranolol in order to investigate the participation of catecholaminergic system as well as the α- and β-adrenergic receptors to the antihyperalgesic effect of atomoxetine, respectively. Obtained findings revealed that AMPT, phentolamine and propranolol pre-treatments antagonize the antihyperalgesic activity of atomoxetine.In this thesis study, it was first demonstrated that atomoxetine exhibits a significant antihyperalgesic activity in diabetic-induced neuropathic pain model and concluded that catecholaminergic system as well as α- and β-adrenergic receptors mediates this effect. These findings point to the possibility of a new indication for atomoxetine.

Selektif noradrenalin geri alım inhibitörü bir ilaç olan atomoksetin, dikkat eksikliği hiperaktivite bozukluğunun tedavisinde endikedir. Beyinde monoamin konsantrasyonunu artırıcı etki gösteren ilaçların nöropatik ağrı tedavisi için terapötik potansiyele sahip olduğu bilgisinden hareketle, bu çalışmada atomoksetin'in diyabet ile indüklenen nöropatik ağrı üzerine olası etkinliğinin araştırılması planlanmıştır.Randall-Selitto ve Hargreaves testlerinden elde edilen veriler diyabetin, sıçanların pençe çekme eşik değerlerinde ve pençe çekme sürelerinde anlamlı azalmaya neden olduğuna işaret etmiştir. Diğer yandan, atomoksetin'in 7 ve 14 gün süre ile 3 mg/kg/gün dozda uygulanması diyabet ile indüklenen söz konusu hiperaljezik yanıtları düzeltmiştir. Bu çalışma kapsamında, atomoksetin'in antihiperaljezik etkisine katekolaminerjik sistemin ve ayrıca α- ve β-adrenerjik reseptörlerin katılımının araştırılması amacıyla sırasıyla α-metil-para-tirozin metil ester (AMPT), fentolamin ve propranolol ile mekanistik çalışmalar yapılmıştır. Elde edilen veriler AMPT, fentolamin ve propranolol ön-uygulamalarının atomoksetin'in antihiperaljezik etkinliğini antagonize ettiğini ortaya koymuştur. Bu tez çalışmasında atomoksetin'in diyabet ile indüklenen nöropatik ağrı modelinde kayda değer bir antihiperaljezik etkinlik gösterdiği ilk kez ortaya konulmuş ve bu etkiye katekolaminerjik sistemin ve α- ve β-adrenerjik reseptörlerin aracılık ettiği sonucuna ulaşılmıştır. Elde edilen bu bulgular atomoksetin için yeni bir endikasyon olasılığına işaret etmektedir.
