Breast cancer, which is the second most common cause of cancer deaths, is one of the problems of global public health since it fail store spend to treatment and has a complex etiology. It is argued that anti estrogenic agents such as melatonin will lead to breast cancer treatment as estrogen has a stimulating effect on growth of breast cancer. In addition, zinc, which plays a crucial role in the etiology of cancer and is vital for many cellular processes, is an important trace mineral. Trace elements are regarded as versatile anti-cancer agents that regulate various biological mechanisms.In this study, it was aimed to investigate the effects of application of melatonin and zinc on lipid peroxidation and elemental metabolism in female rats with DMBA-induced mammary carcinoma. The study groups are composed of control, DMBA control and treatment (zinc, melatonin and Zinc+Melatonin) groups. Female rats (except the control group) were given a strong carcinogen DMBA. After tumor formation, zinc and melatonin were administered to the treatment groups at a dose of 5 mg/kg/day for 4 weeks. Four weeks later, the administration of the test animals was terminated by decapitation, and blood and breast tissue samples were taken. MDA as an indicator of tissue damage and GSH levels as an indicator of antioxidant activity in blood and breast tissue samples were determined by spectrophotometric method. Besides, some serum element levels (iron, magnesium, zinc and copper) were calculated.The highest level of MDA found in plasma and breast tissue was obtained in the DMBA control group (P<0,05). Plasma MDA levels of the breast tissue were lower in the zinc group than in the melatonin group and significantly decreased in the zinc + melatonin group (P<0,05).Plasma MDA levels showed a significant decrease in the Zinc+Melatonin group (P<0,05) although there was no difference in the zinc and melatonin groups. The lowest GSH level found in erythrocytes and breast tissue was obtained in the DMBA control group. When zinc and melatonin groups were compared in the breast tissue, GSH level of zinc group was found to be higher than GSH level of melatonin group (P<0,05). Furthermore, the highest increase was found in the zinc + melatonin group. The GSH levels measured in erythrocyte increased in the zinc + melatonin group (P<0,05), although there was no difference in the zinc and melatonin groups. The highest levels of iron, magnesium and zinc were found in the zinc + melatonin group, and the highest level of copper was obtained in the DMBA control group (P<0,05).The findings showed that the antioxidant activity suppressed by increased oxidative damage in rat mammary carcinoma was improved, but the administration of Zinc + Melatonin gave better results.

Kanser ölümlerinin ikinci en yaygın nedeni olan meme kanseri, tedaviye yetersiz cevap vermesi ve karmaşık etiyolojiye sahip olmasından dolayı küresel halk sağlığı problemlerindendir. Meme kanseri gelişmesinde, östrojenin uyarıcı etkiye sahip olması nedeniyle, melatonin gibi anti östrojenik ajanların meme kanseri tedavisine yön vereceği ileri sürülmektedir. Ayrıca çinko ise, çok sayıdaki hücresel işleyiş için yaşamsal önemi olan ve kanser etyolojisinde önemli rol oynayabilen bir iz mineralidir. Eser elementler, çeşitli biyolojik mekanizmaları düzenleyen çok yönlü kanser önleyici maddeler olarak kabul edilmektedir.Bu çalışmada, DMBA ile meme kanseri oluşturulmuş dişi sıçanlarda çinko ile melatonin uygulamasının lipit peroksidasyonu ve element metabolizmasına olan etkilerinin araştırılması amaçlanmıştır. Çalışma gruplarını, kontrol, DMBA kontrol ve tedavi (çinko, melatonin ve Çinko+Melatonin) grupları oluşturmaktadır. Dişi sıçanlara (kontrol grup hariç) güçlü bir kanserojen madde olan DMBA verilmiştir. Tümör oluştuktan sonra, tedavi gruplarına 5 mg/kg/gün dozda çinko ve melatonin 4 hafta süre ile uygulanmıştır. Dört hafta sonra deney hayvanları uygulaması dekapitasyonla sonlandırılmış ve kan ve meme dokusu örnekleri alınmıştır. Alınan kan ve meme doku örneklerinde doku hasarının göstergesi olarak MDA, antioksidan aktivitenin bir göstergesi olarak da GSH düzeyleri spektrofotometrik yöntemle tayin edilmiştir. Ayrıca bazı serum element düzeyleri (demir, magnezyum, çinko ve bakır) belirlenmiştir.Plazma ve meme dokusunda belirlenen en yüksek MDA düzeyi DMBA kontrol grubunda elde edilmiştir (P<0,05). Meme dokusu plazma MDA düzeyleri çinko grubunda, melatonin grubuna göre düşük ve Çinko+Melatonin grubunda ise anlamlı derecede azalmıştır (P<0,05). Plazma MDA düzeylerinde ise çinko ve melatonin gruplarında fark olmamasına rağmen, Çinko+Melatonin grubunda anlamlı derecede azalma belirlenmiştir (P<0,05). Eritrosit ve meme dokusunda belirlenen en düşük GSH düzeyi DMBA kontrol grubunda elde edilmiştir. Meme dokusunda çinko ve melatonin grupları kıyaslandığında çinko grubunun GSH düzeyi, melatonin grubunun GSH düzeyine göre daha yüksek olduğu belirlenmiştir (P<0,05). Ayrıca, en fazla artış Çinko+Melatonin grubunda belirlenmiştir. Eritrositte ölçülen GSH düzeylerinde çinko ve melatonin grubunda fark olmamasına rağmen, Çinko+Melatonin grubunda yükselmiştir (P<0,05). Demir, magnezyum ve çinkonun en yüksek düzeyleri Çinko+Melatonin grubunda, bakırın en yüksek düzeyi ise DMBA kontrol grubunda elde edilmiştir (P<0,05). Elde edilen bulgular, sıçanlardaki meme kanserinde artan oksidan hasarın artması ile baskılanan antioksidan aktivitenin, çinko ve melatonin uygulaması ile düzeltildiği, ancak Çinko+Melatonin uygulamasının daha iyi bir şekilde sonuç verdiğini göstermektedir.
