In this study, it was aimed to investigate the protective effects of 6-shogoal and zingerone in Zingiber officinale (ginger) and to compare these effects with cucumin, a potent antioxidant, in an experimental kidney injury model generated with cisplatin in rats. Cisplatin is an antineoplastic agent used in the treatment of many cancer types. The side effects of cisplatin, especially the dose-dependent nephrotoxic effect, are the major factors that limit its use in cancer treatment. Due to the resulting kidney damage, precautions to reduce the adverse effects of this drug are needed. Ginger contains non-volatile bioactive molecules such as gingerol, shogaol and zingerone. The antioxidant, antimigrenic, antiemetic and antilipidemic effects of these substances are demonstrated by in vivo and in vitro models.Sixtyfour Wistar Albino rats were used in the study. Experiment were performed on 8 groups; control, cisplatin, 6-shogaol, zingerone, curcumin, cisplatin + 6-shogaol, cisplatin + zingerone, cisplatin + curcumin group. Nephrotoxicity was induced by administering a single dose of 7 mg / kg cisplatin (ip) on the 10th day of drug administration to the animals. The treatment groups received 10 mg / kg / day 6-shogaol, 6 mg / kg / day zingerone and 200 mg / kg / day curcumin by oral gavage for 14 days.BUN and creatinine levels, MDA levels and SOD and GSHPx enzyme activities in serum and kidney tissues were measured and histopathologic examination was performed in kidney tissues.BUN, creatinine and MDA levels increased (p <0.01) and SOD and GSHPx enzyme levels decreased in cisplatin group (p <0.01). 6-shogaol and zingerone treatment groups showed a decrease in BUN, creatinine and MDA levels when compared to the cisplatin group (p <0.05). GSHPx enzyme levels increased in the 6-shogaol group (p <0.05). In the zingeron treatment group, both SOD and GSHPx enzyme levels increased (p <0.05). When the treatment group results were compared with the curcumin group, similar results were obtained especially in the zingeron group. Histopathological findings supported biochemical findings.In conclusion, the effects of 6-shogaol and zingerone on nephrotoxicity induced by cisplatin in rats were investigated for the first time. In this study, it was concluded that 6-shogaol and zingerone have protective effects against cisplatin nephrotoxicity and that this protective effect may be partially due to antioxidant properties of 6-shogaol and zingerone.Key words: Cisplatin, Nephrotoxicity, 6-shogaol, Zingerone, Curcumin, Rat.

Bu çalışmada, sıçanlarda sisplatin ile oluşturulan deneysel böbrek hasarı modelinde Zingiber officinale'de (zencefil) bulunan 6-shogoal ve zingeron'un koruyucu etkilerinin araştırılması ve bu etkilerin güçlü bir antioksidan olan kurkuminle karşılaştırılması amaçlandı. Sisplatin pek çok kanser türünün tedavisinde kullanılan bir antineoplastik ajandır. Sisplatinin yan etkileri, özellikle doza bağımlı nefrotoksik etkisi kanser tedavisinde kullanımını kısıtlayan majör faktördür. Oluşan böbrek hasarı nedeniyle, bu ilacın istenmeyen etkilerini azaltıcı önlemler alınmasına ihtiyaç duyulmaktadır. Zencefilin içeriğinde gingerol, shogaol ve zingeron gibi uçucu olmayan biyoaktif moleküller bulunur. Bu maddelerin antioksidan, antimigren, antiemetik ve antilipidemik etkileri in vivo ve in vitro deney modelleriyle ortaya konulmuştur. Çalışmada 64 adet Wistar Albino ırkı sıçan kullanıldı. Deneyler; kontrol, sisplatin, 6-shogaol, zingeron, kurkumin, sisplatin+6-shogaol, sisplatin+zingeron, sisplatin+kurkumin olmak üzere 8 grup üzerinde gerçekleştirildi. Nefrotoksisite hayvanlara ilaç uygulamalarının 10. günü tek doz 7mg/kg sisplatin (i.p) uygulanarak oluşturuldu ve tedavi gruplarına 14 gün süre ile oral gavajla, 10 mg/kg/gün 6-shogaol, 6 mg/kg/gün zingeron ve 200 mg/kg/gün kurkumin uygulandı.Alınan serum örnekleri ve böbrek dokularında BUN ve kreatinin düzeyleri, MDA düzeyleri ile SOD ve GSHPx enzim aktiviteleri ölçüldü, ayrıca böbrek dokularında histopatolojik inceleme yapıldı. Sisplatin uygulanan grupta BUN, kreatinin ve MDA düzeyleri artarken (p<0.01), SOD ve GSHPx enzim düzeylerinde azalma meydana geldi (p<0.01). 6-shogaol ve zingeron tedavi grupları sisplatin grubu ile karşılaştırıldığında BUN, kreatinin ve MDA düzeylerinde azalma gözlendi (p<0.05). 6-shogaol grubunda GSHPx enzim düzeylerinde artış meydana geldi (p<0.05). Zingeron tedavi grubunda hem SOD ve GSHPx enzim düzeylerinde artış tespit edildi (p<0.05). Tedavi grubu sonuçları kurkumin grubu ile karşılaştırıldığında özellikle zingeron grubunda benzer sonuçlar elde edildi. Histopatolojik bulgular biyokimyasal bulguları destekledi. Sonuç olarak 6-shogaol ve zingeron'nun, sıçanlarda sisplatinle ile oluşturulan nefrotoksisiteye karşı etkilerinin ilk defa araştırıldığı bu çalışmada, 6-shogaol ve zingeronun sisplatin nefrotoksisitesine karşı koruyucu etkileri olduğu ve bu koruyucu etkinin kısmen 6-shogaol ve zingeron'nun antioksidan özelliklerine bağlı olabileceği sonucuna varılmıştır.
