Cancer is a disease which its incidence is raising in the world and also hard to curative treatment yet. Excessive of spreading of the disease and low probability of definitive treatment causes to increase the importance of early diagnosis and detection. In diagnosis period, histochemical evaluation is a significant step. Tumor staging and type of damage on tissue injury can be done with analyzing histopathologic image. Beside tumor staging, to determine the rate of proliferation of cell allows to getting information about the course of disease and choosing the more appropriate treatment method.This research aims to support and help pathologists within the cancer diagnosis and detection. In this process, images on cancerous tissue which colored by Kİ-67, stained cells are automatically segmented and total number of cells is calculated. The calculated number is one of the parameters used to define the proliferation speed of tumor cells. In order to calculate the positive cell numbers in histopathologic images, a successive two stage algorithm is used that determine separate and touching cells. At the first stage, designed two-dimensional optimal filter is used to clear background data on images and separate cells are detected on these images by using morphological and edge-detection operations. At the second stage, Level-Set Algorithm is used to segment touching cells. At last stage, all positive cells are calculated by determining.This study aims to contribute knowledge the level of Biomedical Engineering area of Turkey, increase the decision support mechanism for doctors and the last but not the least to find a solution to medical problems with engineering approach.

Kanser, dünya genelinde hızla yayılan ve henüz kesin tedavisi olmayan bir hastalıktır. Hastalığın oldukça sık görülmesi ve kesin tedavisinin zor olması, erken tanı ve teşhisin önemini artırmaktadır. Tanı aşamasında histokimyasal değerlendirme önemli bir basamaktır. Histopatolojik görüntülerin incelenmesi ile dokularda oluşan hasarın çeşidi belirlenerek tümör evrelendirmesi yapılmaktadır. Ayrıca, tümör evrelendirmesi ile birlikte hücrenin proliferasyon hızının belirlenmesi hastalığın seyri hakkında bilgi edinilerek en uygun tedavi yönteminin seçimine olanak sağlanır. Bu çalışmada kanser hastalığının tanı ve teşhisinde patologlara yardım ve destek olması amaçlanmıştır. Kanserli dokulara ait Ki-67 boyama ile elde görüntülerde boyanan hücreler otomatik olarak bölütlenmiş ve toplam sayısı hesaplanmıştır. Hesaplanan bu sayı tümörlerin hücre çoğalma hızını belirlemede kullanılan parametrelerden biridir. Histolopatolojik görüntülerdeki pozitif hücrelerin sayısının hesaplanmasında kesişen ve kesişmeyen hücrelerin belirlendiği ardışık iki aşamalı bir algoritma kullanılmıştır. İlk aşamada tasarlanan iki boyutlu optimal filtre ile arka plan verilerinden temizlenmiş, bu görüntüye morfolojik işlemler ve kenar belirleme işlemi uygulanarak kesişmeyen hücreler belirlenmiştir. İkinci aşamada kesişen hücrelerin bölütlenmesi için seviye küme algoritması kullanılmıştır. Sonuç olarak pozitif hücreler belirlenerek sayımı gerçekleştirilmiştir.Bu çalışma ile ülkemizin biyomedikal mühendisliği alanında bilgi düzeyine katkıda bulunulması ve hekime karar destek mekanizmasının artırılması ile tıpta karşılaşılan problemlere mühendislik yaklaşımı ile çözüm amaçlanmıştır.
