High-fructose intake in the diet can cause metabolic syndrome (MetS). Oxidative stress and inflammatory variables caused by the consumption of fructose impair the kidney function and increase the risk of disease. Probiotics might have antioxidant, antiinflammatory and immunoregulatory properties. However, there was limited data on the influence of probiotics treatment in high-fructose-induced metabolic syndrome. The present study was aimed to search the influence of the supplementation of Lactobacillus helveticus (L. helveticus) and Lactobacillus plantarum (L. plantarum) on dietary fructose-induced metabolic changes and renal antioxidant/oxidant status of rats. Male Wistar rats were randomly divided into four groups as control; fructose; fructose+L. helveticus and fructose+L. plantarum. Fructose was given to the rats as 20% solution in drinking water for 15 weeks and the probiotics supplementation (10+9 CFU/ml/100 g) was applied by gastric gavage once a day during 6 weeks from the 9th week of the application of fructose. Glucose, insulin, triglyceride, VLDL, HDL, total cholesterol, MDA, creatinine, urea, uric acid, albumin, and total protein in the samples of plasma with renal MDA, urea, and uric acid levels were measured by biochemical methods. The main antioxidant enzymes SOD1, SOD2, CAT, and GPx mRNA/protein expressions in kidney tissues were determined by qRT-PCR and Western Blot analysis methods. Dietary fructose-induced elevations in plasma insulin, triglyceride, VLDL, MDA, and creatinine as well as renal MDA, and urea levels were alleviated following the treatment with L. helveticus, and L. plantarum. Moreover, L. helveticus, and L. plantarum supplementation importantly recovered the changes in renal protein expression level of SOD1, SOD2 and CAT. In conclusion, the supplementation of L. helveticus, and L. plantarum has an improving effect on certain metabolic parameters and renal antioxidative enzymes in fructose-induced MetS.

Diyet içerisinde yüksek fruktoz alımı metabolik sendroma (MetS) neden olabilir. Fruktoz tüketiminin neden olduğu oksidatif stres ve inflamatuvar değişkenler ise böbrek fonksiyonlarını bozarak hastalık riskini artırır. Probiyotikler antioksidan, antiinflamatuvar ve immünomodülatör etkilere sahip bakterilerdir. Literatürde, probiyotiklerin yüksek fruktoz diyeti ile oluşturulan MetS modelleri üzerine etkilerini inceleyen sınırlı çalışma yer almaktadır. Bu çalışmada, liyofilize Lactobacillus helveticus (L. helveticus) ve Lactobacillus plantarum (L. plantarum) suşlarının yüksek fruktozun neden olduğu metabolik değişiklikler ve renal oksidatif stres üzerindeki etkileri araştırılmıştır. Wistar erkek sıçanlar dört gruba ayrılmıştır; kontrol, fruktoz, fruktoz+L.helveticus ve fruktoz+L. plantarum. Fruktoz 15 hafta süre ile içme suyu içerisinde %20 çözelti şeklinde verilirken; probiyotikler ise fruktoz uygulamasının 9.haftasından itibaren 6 hafta boyunca günde bir kez (10+9CFU/ml/100 g) olmak üzere gastrik gavaj ile uygulanmıştır. Sıçanların plazma örneklerinde glukoz, insülin, trigliserit, VLDL, HDL, total kolesterol, MDA, kreatinin, üre, ürik asit, albümin ve total protein ile renal MDA, üre ve ürik asit düzeyleri biyokimyasal yöntemlerle ölçülmüştür. Böbrek dokularındaki temel antioksidan enzimler olan SOD1, SOD2, CAT ve GPx mRNA/protein ekspresyonları ise qRT-PCR ve Western Blot analiz yöntemleri ile tespit edilmiştir. Fruktoz tüketimiyle artan plazma insülin, trigliserit, VLDL, MDA ve kreatinin ile renal MDA ve üre düzeyleri üzerine L. helveticus ve L. plantarum uygulamasının hafifletici etkileri tespit edilmiştir. Diğer taraftan, L. helveticus ve L. plantarum takviyesiyle renal SOD1, SOD2 ve CAT protein ekspresyon seviyesindeki değişiklikler önemli ölçüde geri kazanılmıştır. Sonuç olarak, diyet içerisinde L. helveticus ve L. plantarum alımının, fruktozun neden olduğu MetS'da belirli metabolik parametreler ve renal antioksidan enzimler üzerinde iyileştirici bir etkisi olduğu söylenebilir.
