Atherosclerosis is a chronic inflammatory disease, which the cellular and humoral immune responses are included, caused by the constriction of the vessel lumen because of the atherosclerotic plaques as a result of the thickening and hardening of medium and larger arteries. The cells going through the atherosclerotic process secrete cytokines and they also have the ability of being activated by the cytokines. It is demonstrated that inflammation generates accumulation of the monocytes and/or lymphocytes as a result of the expression of adhesion molecules, cytokines and chemokines (MCP-1, IL-8, ICAM-1, VCAM-1, E-selectin and P-selectin) after activation of the endothelium, which consequently results in infiltration of the subendothelium. Anacardic acid (AA) is a bioactive phytochemical found in the nutshell of the Anacardium occidentale (cashew apple). AA may provide better protection against several pathophysiological disorders, such as oxidative damage or cancer. Several studies on NF-κB activation pathways by using AA and on apoptosis and inflammation mechanisms of NF-κB regulatory gene products suggest that AA suppresses NF-κB, which is activated by the growth factors and inflammatory responses.In our study, we aimed to investigate the relation between AA and ICAM-1, VCAM-1 and NF-κB, which are associated with atherosclerotic process and with formation of atheroma plaque, by using the inflammation model of human saphenous vein endothelial culture constituted by TNF-α stimulation.In our study, the acute effect of the AA on adhesion molecules and on NF-κB has been determined by applying TNF-α on endothelial cells after acute application of AA. Afterwards, changes in mRNA levels of ICAM-1, VCAM-1 and NF-κB genes have been determined by using qRT-PCR method. Double immunofluorescence procedure has been applied to NF-κB and to ICAM-1 proteins. The cytotocixity test has been applied to all groups. As a consequence of our study, it is considered that AA can be effective on prevention of the development of saphenous vein graft disease and on its treatment by altering the expression of adhesion molecules through NF-κB pathway.

Ateroskleroz, hücresel ve hümoral immün yanıtların katıldığı, orta ve daha büyük boyutlardaki arterlerin kalınlaşması ve sertleşmesi sonucunda damar lümeninin aterosklerotik plaklarla daralması ile oluşan kronik inflamatuar bir hastalık olarak tanımlanmaktadır. Aterosklerotik süreçte yer alan hücreler, sitokinleri salgıladıkları gibi, sitokinler tarafından da aktive edilebilme özelliğine sahiptirler. İnflamasyonun, endotel aktive olduktan sonra adezyon moleküllerinin, sitokinlerin ve kemokinlerin (MCP-1, IL-8, ICAM-1, VCAM-1, E-selektin ve P-selektin) ekspresyonuyla monositlerin ve/veya lenfositlerin toplanmasına ve subendotelyumda infiltrasyona yol açtığı bildirilmiştir. Anakardik asit (AA), Anacardium occidentale (kaju fıstığı) kabuğunda bulunan biyoaktif bir fitokimyasal maddedir. AA, oksidatif hasardan kansere kadar birçok patofizyolojik bozukluk için iyi bir koruma sağlayabilir. AA kullanılarak NF-κB aktivasyon yolağı ve NF-κB düzenleyici gen ürünlerinin apoptoz ve inflamasyon mekanizmaları üzerine yapılan çalışmalarda AA'nın, büyüme faktörleri ve inflamatuar yanıtlarla aktiflenen NF-κB yi baskıladığı bildirilmiştir. Çalışmamızda, AA'nın insan safen ven endotel kültüründe, TNF-α ile uyarımı sonucu oluşturulan inflamasyon modelinde ICAM-1, VCAM-1 ve NF-κB arasındaki ilişkiyi araştırmayı amaçladık. Çalışmamızda endotel hücreleri üzerine kısa süreli AA uygulamasının ardından TNF-α uygulaması yapılarak, AA'in adezyon molekülleri ve NF-κB üzerine olan akut etkisi saptanmıştır. Daha sonra gerçek zamanlı kantitatif polimeraz zincir reaksiyonu yöntemi ile ICAM-1, VCAM-1 ve NF-κB genlerinin mRNA seviyelerindeki değişiklikler saptanmıştır. NF-κB ve ICAM-1 proteinleri için ise ikili immünfluoresan boyama prosedürü uygulandı. Tüm gruplara sitotoksisite testi uygulandı. Çalışmamızın sonuçlarına göre AA'nın, safen ven greft hastalığının gelişmesinin önlenmesinde ve tedavisinde NF-κB yolağı üzerinden, adezyon moleküllerinin ekspresyonunu etkileyerek etkin olabileceği düşünülebilir.
