ABSTRACTInvestigation Of The Protective Effects Of Ellagic Acid In The Prevention Of The Kidneys Damage Occurring In Rats Exposed To Nicotine In Fetal PeriodAim: In this study, the protective effects of ellagic acid in the prevention of the kidneys damage occurring in rats exposed to nicotine in fetal period were investigatedMaterials and method: In this study, 20 admit female rats were used.The rats were divided into 4 groups as 5 rats were in one group. The group were Control, Nicotine, Ellagic acid, Nicotine + Ellagic acid. A male rat was put into each cage (group) so that the female rats can pregnant. Nicotine and ellagic acid treatments continued throughout the pregnant period and also for 15 days after deliver. At the end of 15 th day all neonatal rats were killed under ether anaesthesia and their kidneys were taken for biochemically and histopathologically examinations.Results: Lower body weight, total glutathione (GSH) level, the activities of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD), and higher levels of malaondialdehyde (MDA) and nitric oxide (NO) were found in neonatal rats of female mothers which were exposed to nicotine, compared with the control group. In addition, attenuation of the development of the kidneys was determined. On the other hand these values were similar to control group when the values of nicotine+ ellagic acid group were compared with nicotine group.Conclusion: As a result, the existence of kidney damage in neonatal rats of the nicotine treated pregnant rats was confirmed by both biochemical (increased MDA and NO levels and decreased GSH, GSH-Px and SOD levels compared with control group) and immunohistochemical. It was detected that by means of ellagic acid application of histological changes and oxidative stress occurring by nicotine would be reduced.Key Words: Kidney, Ellagic acid, Fetus, Nicotine.

ÖZETFötal Dönemde Nikotine Maruz Kalan Sıçanlarda Oluşan Böbrek Hasarının Engellenmesinde Ellagic Asitin Koruyucu Etkilerinin İncelenmesiAmaç: Bu çalışmada, fötal dönemde nikotine maruz bırakılan sıçanlarda oluşan böbrek hasarının engellenmesinde ellagic asitin koruyucu etkisi incelendi.Materyal ve metot: Bu çalışmada yetişkin 20 adet dişi rat kullanıldı. Ratlar her grupta 5 adet olacak şekilde Kontrol, Nikotin, Ellagic asit, Nikotin + Ellagic asit olarak 4 gruba ayrıldı. Her kafese 1 adet erkek rat bırakılarak ratların gebe kalması sağlandı. Nikotin ve Ellagic asit uygulaması gebelik boyunca ve doğumu izleyen 15. güne kadar devam ettirildi. Onbeşinci günün sonunda bütün gruplardaki yavrular eter anestezisi altında ötenazi edilerek böbrek dokuları biyokimyasal ve histopatolojik incelemeler için alındı.Bulgular: Yapılan değerlendirmelerde kontrol grubu ile karşılaştırıldığında nikotin uygulanan annelere ait yavruların vücut ağırlığı, (GSH), (GSH-Px) ve (SOD) enzim düzeylerinde azalma, (MDA) ve (NO) düzeylerinde ise artış görüldü. Ayrıca histokimyasal incelemelerde böbrek gelişiminin yavaşladığı'da tespit edildi.Diğer taraftan Nikotin + Ellagic asit verilen gruba ait yavrulardaki bu değerler sadece nikotin verilen grup ile karşılaştırıldığında değerlerin kontrol grubuna yakın olduğu görüldü.Sonuç: Sonuç olarak, nikotin uygulanan gebe ratların yavrularına ait böbrek dokularındaki hasarın varlığı hem biyokimyasal olarak (kontrol grubuna göre MDA ve NO düzeylerinde artış, GSH, GSH-Px ve SOD gibi antioksidan enzim düzeylerinde ise düşüş hem de immunhistokimyasal olarak tespit edilmiştir. Ellagic asit uygulaması ile nikotinin böbrekte oluşturduğu oksidatif stres ve histolojik değişikliklerin azaltılabileceği belirlenmiştir.Anahtar Kelimeler: Böbrek , Ellagic asit, Fötüs, Nikotin.
