Yoghurt, which is a traditional product widely consumed with different varieties in ourcountry and in the world, is important for healthy nutrition. However, most of thebacteria originated from yoghurt can not pass through the stomach and intestinalbarriers and this situation cause some problems on probiotic effect of the product. Theaim of this study is to isolate probiotic strains from traditional yoghurt samples, whichcan pass through stomach and bile barriers. For this purpose, some microbiological andphysicochemical analyzes have been applied on a total of 50 traditional and industrialyoghurt samples. As a result of the analyzes, the means of probable lactic bacilli andcocci, yeast-mold, coliform, pH and acidity (lactic acid) values of the samples were6.56; 4.96; 2.19; <1; 3.74 and 1.17%, respectively. Strain-based differences of isolateswere determined by using (GTG)5 fingerprint profiles. 11 different strains in total 198isolates were determined by moleculer techniques. It was observed that 24.2% of the198 strains isolated in the study were able to pass the stomach and bile barriers and hadprobiotic efficacy. The isolates were determined as 4 different Lactobacillus delbrueckiisubsp. bulgaricus and 7 different Streptococcus salivarius subsp. termophilus strains.Keywords: yoghurt, probiotic, GTG5 fingerprinting.

Geleneksel bir ürünümüz olan, ülkemizde ve dünyada farklı çeşitleri ile yaygın olaraktüketilen yoğurdun mikrobiyotası sağlıklı beslenme açısından önem arz etmektedir.Ancak yoğurdu oluşturan bakterilerin büyük çoğunluğunun mide ve bağırsakbariyerlerini geçememesi probiyotik etkisi ile ilgili bazı soru işaretleri doğurmaktadır.Bu çalışmada geleneksel olarak üretilen yoğurt örneklerinden mide ve safra bariyerlerinigeçebilen muhtemel probiyotik suşların izolasyonu amaçlanmaktadır. Bu amaçlageleneksel ve endüstriyel olarak üretilmiş toplam 50 yoğurt örneği üzerinde bazımikrobiyolojik ve fizikokimyasal analizler gerçekleştirilmiştir. Yapılan analizler sonucuörneklere ait ortalama muhtemel laktik basil, muhtemel laktik kok, maya-küf, koliform,pH ve % asitlik (laktik asit) miktarları sırası ile 6,56; 4,96; 2,19; <1; 3,74 ve %1,17bulunmuştur. İzolatların suş bazında farklılıklarının ortaya koyulması amacı ile (GTG)5parmak izi profilleri elde edilmiştir. Moleküler seviyede 11 farklı suşun varlığıbelirlenmiştir. Çalışmada elde edilen toplam 198 suşun %24,2'sinin mide ve safrabariyerini geçebildiği ve ilgili probiyotik yeterliğe sahip olduğu görülmüştür.
