ABSTRACTThis research aimed to investigate the effects of methylglyoxal, which is a metabolic end product in the cell due to high fructose diet consumption, on beta-amyloid formation in vitro, secondary messenger molecule cyclic adenosin monophosphate (cAMP) and its effect on phosphoesterase 1 (PDE1), which degrades this molecule.Human glioblastoma U-87 MG (ATCC HTB-14) cell line was used in the study. Cells were maintained in a sterile incubator at 37 ° C, 5% CO 2 and 95% air in Eagle's Minimum Essential Medium (E-MEM) medium containing 25% fetal bovine serum and 10 % fetal bovine serum (FBS) and penicillin / streptomycin / amphotericin B mixture. Methylglyoxal was added at different concentrations (100-400 μM) to the cells cultured in 24-well flask one day before. At the end of the period, the effective concentration of methylglyoxal was determined by adding MTT reagent to the wells. In subsequent experiments, this concentration was applied to the cells, and the expression levels of PDE 1B, PDE 1B1 and Aβ1-40 gene from the RNA samples taken at the end of 24 hours were investigated using real time PCR method. Again, cAMP levels of the protein samples were analyzed by ELISA method. At the end of the incubation period, groups of cells were photographed for amyloid aggregation images with cell density and Congo red staining.In the viability tests performed in the study, it was observed that the concentration of 200 μM of methylglyoxal decreased the cells by 28% compared to the control group. This concentration was taken as the effective concentration and used for further studies. Again, it was determined that methylglyoxal increased the cellular cAMP level by 31%. According to the obtained gene expression data, methylglyoxal increased the Aβ1-40 gene expression 3.7-fold, while the PDE1B and PDE1B1 gene expressions were downregulated 1.6 and 3.3-fold, respectively. In the study, it was observed that as a result of the Congo red dyeing, the centers of the methylglyoxal-added cells were surrounded by blue color and the peripheries were surrounded by yellow-colored condensed amyloid aggregatesIn the present thesis, it was concluded that the fructose diet would significantly affect the pathogenesis of Alzheimer's disease in affected individuals. However, in order to fully illuminate the subject, further research is needed, especially in vivo experiments.Key words: Alzheimer, cAMP, phosphodiesterase, amyloid protein.

Araştırmada yüksek früktoz diyeti tüketimine bağlı hücrede ortaya çıkan metabolik son ürün metilgliyoksalin in vitro beta amiloid oluşumu, ikincil haberci molekül siklik adenozin monofosfat (cAMP) ve bu molekülün parçalanmasını sağlayan fosfodiesteraz 1 (PDE1) üzerine etkilerinin araştırılması amaçlandı. Çalışmada insan orijinli glioblastoma U-87 MG (ATCC HTB-14) hücre hattı kullanıldı. Hücreler, 25 cm2 lik flasklarda ve içeriğinde %10 oranında fötal sığır serumu (FBS) ve penisilin/ streptomisin/amfoterisin B karışımı bulunduran Eagle's Minimum Essential Medium (E-MEM) besi ortamında, 37oC, %5 CO2 ve %95 hava bulunduran steril inkübatörde üretildi. Çalışmada 1 gün önceden 24 kuyucuklu besi kaplarına ekilen hücrelere farklı konsantrasyonlarda metilgliyoksal(100- 400 um) eklendi. Süre sonunda kuyucuklara mtt ayıracı eklenerek metilgliyoksalin etkin konsantrasyonu tespit edildi. Sonraki denemelerde bu konsantrasyon hücrelere uygulanarak 24 saatin sonunda alınan RNA örneklerinden PDE 1B, PDE 1B1 ve Aβ1-40 gen ekspreyon düzeyleri real time PCR metoduyla araştırıldı. Yine alınan protein örneklerinden cAMP düzeyi ise ELİSA metoduyla analiz edildi. İnkübasyon süresi sonunda gruplardan hücre yoğunluk ve Kongo red boyaması ile amiloid birikim görüntüleri fotoğraflandı. Araştırmada yapılan canlılık kontrollerinde metilgliyoksalin 200 uM konsantrasyonu kontrol grubuna göre hücreleri %28 oranında azalttığı gözlendi. Bu konsantrasyon etkin konsantrasyon olarak kabul edilerek sonraki çalışmalar için kullanıldı. Yine metilglyoksalin hücreiçi cAMP düzeyini %31 oranında arttırdığı tespit edildi. Elde edilen gen ekspresyon verilerine göre metilgliyoksal Aβ1-40 gen ekspresyonunu 3.7 kat arttırıken, sırasıyla PDE1B ve PDE1B1 gen ekspresyonlarını ise 1.6 ve 3.3 kat baskıladığı ettiği tespit edildi. Çalışmada Kongo red boyaması sonucunda metilgliyoksal eklenen hücrelerin merkezlerinin mavi renkte ve periferlerinde ise sarı, yoğunlaşmış amiloid birikimlerinin olduğu gözlendi. Yapılan araştırmada fruktoz diyetinin etkilenen bireylerde Alzheimer hastalığının patogenezinde anlamlı derecede etkileyeceği kanısına varıldı. Ancak konunun tam aydınlatılması için başta in vivo denemeler olmak üzere ve daha ileri araştırmalara ihtiyaç duyulmaktadır. Anahtar kelimeler: Alzheimer, cAMP, fosfodiesteraz, amiloid protein.
