Amanita genus has poisonous species that are so called death cap or death angel which are responsible for more than 90% of mushroom poisoning as well as having few edible species. Species in the genus Terfezia have a high nutritional value and economic importance. Morphological identification of these species is time consuming and can be misleading and experience is required. Also results are subjective, sometimes showing inconsistencies among the researchers. In this study Amanita species from Turkey, and USA, and Terfezia species from different region of Turkey, and Syria were studied. The aim of this study was to perform molecular identification of these Amanita and Terfezia species and determine whether they have any polymorphism at their ITS nuclear ribosomal DNA sequences.. To achieve this goal, sequences of internal transcribed spacers (ITS) region in the nuclear rDNA were amplified with PCR (Polymerase Chain Reaction) technique by using ITS1, ITS4 primers. PCR products were sent for sequence analysis. Sequences from analysis were aligned with BioEdit biological sequence alignment editor software that was designed for Windows 95/98/NT/2000/XP. Obtained sequences were compared with GeneBank database by using BLAST program. Phylogenetic and phenetic trees were produced from the sequences by using Neighbour joining, Branch and Bound, UPGMA and Bootstrap analysis methods based on PAUP* software. As result, Amanita species?s morphologic diagnosis were verified also by molecular techniques and the first sequencing of ITS region of Amanita canescens and Amanita jacksonii was performed. This information will be deposited in the GenBank (NCBI). Polymorphisms were determined in the sequences of Amanita bisporigera, Amanita brunnescens and Amanita caesarea. All Terfezia samples were identified as Terfezia claveryi. A single nucleotide polymorphism was determined in DNA fragments of Terfezia samples from Elazığ in Turkey.Keywords: Amanita spp., Terfezia spp., Internal Transcribed Spacer (ITS).

Amanita cinsi az sayıda yenilebilir türlerinin dışında,`ölüm şapkası ya da ölüm meleği' diye isimlendirilen ve mantar zehirlenmelerinin %90'dan fazlasının sorumlusu olan zehirli türlere sahiptir. Terfezia cinsine ait türler `yer mantarı' diye isimlendirilen ve yüksek besin değerine ve ekonomik öneme sahip türlerdir. Bu türlerin morfolojik tanımlanması zaman alıcıdır, sonuçlar yanıltıcı olabilir ve tecrübe gerektirmektedir. Ayrıca sonuçlar özneldir ve zaman zaman araştırmacılar arasında tutarsızlık göstermektedir. Bu çalışmada, Türkiye ve ABD'den Amanita türleri ile Türkiye'nin değişik bölgelerinden ve Suriye'den elde edilen Terfezia türleri çalışılmıştır. Bu çalışmanın amacı; Amanita ve Terfezia türlerinin moleküler düzeyde tanımlamasını yapmak ve nükleer rDNA'nın tekrarlı birimleri olan ITS dizilerinde herhangi bir polimorfizm olup olmadığını tespit etmektir. Bu amaçla nükleer rDNA'nın tekrarlı üniteleri olan ITS dizileri polimeraz zincir reaksiyonu (PZR) ile, ITS1, ITS4 primerleri kullanılarak çoğaltıldı. PZR ürünleri dizilemeye gönderildi. Gelen diziler Windows 95/98/NT/2000/XP için yazılmış olan BioEdit biyolojik dizi sıralama editörü kullanılarak sıralandı. Elde edilen diziler Genbank'ta BLAST yapılarak GenBank veritabanı sonuçları ile karşılaştırıldı. PAUP programında bulunan Neighbour joining, Branch and Bound, UPGMA ve Bootstrap analiz metotları kullanılarak dizilerin filogenetik ağaçları oluşturuldu. Sonuç olarak, Amanita türlerinin morfolojik teşhisleri moleküler tekniklerle de doğrulandı ve Amanita canescens türünün ITS bölgesi dizisi ilk defa sekanslanmıştır. Bu bilgi genbankasında depolanacaktır. Amanita bisporigera, Amanita brunnescens ve Amanita caesarea'nın dizilerinde polimorfizmler tespit edilmiştir. Terfezia örneklerinin tümü Terfezia claveryi olarak teşhis edilmiş olup Elazığ'dan elde edilen Terfezia örneklerinin dizisinde bir adet single nükleotid polimorfizmi (SNP) tespit edilmiştir.Anahtar Kelimeler: Amanita spp., Terfezia spp., Internal Transcribed Spacer (ITS).
