Introduction and Purpose: The aim of this study is to detect the expression levels of Lipokalin-2 (NGAL- neutrophile gelatinase associated lipokalin) in human prostate cancer tissues having different Gleason score, and to investigate whether there is a relationship between their expression levels and clinicopathologic parameters including response to docetaxel treatment, Gleason score, progression free survival and overall survival.Material-Method: Thirty three patients who received at least three cycles of docetaxel for metastatic castration resistant prostate cancer and whose clinical results were followed were included in the study. Paraffin blocks of the patients were obtained retrospectively from the pathology archives of Çanakkale Onsekiz Mart University Medical Faculty hospital and Çanakkale Mehmet Akif Ersoy Public Hospital. Lipocalin-2 expression status was determined by immunohistochemical (IHC) method using anti-Lipocalin-2 antibody.Findings: Lipokalin-2 was expressed approximately 37% of the study group. The positivity of Lipokalin-2 expression was significantly associated with higher Gleason score (P=0.027). There was no significant association between the expression levels of Lipokalin-2 and docetaxel response. In the group with positive lipocalin-2 expression, the overall survival and progression-free survival times of the patients were numerically shorter, but no statistically significance was found. In this clinicopathological study, findings were confirmed the negative effect of high Lipokalin-2 expression observed in discrete cancer types to prognosis.Conclusion: Our data indicate that high Lipokalin-2 expression might be correlated with poor prognosis in prostate cancer due to its relationship with high Gleason score and short survival. In conclusion, this study supports that Lipocalin-2 expression may be a promising prognostic marker rather than a predictive marker in prostate cancer.Keywords: Prostate adenocarcinoma, Docetaxel, Lipocalin-2, Gleason score, Predictive and Prognostic Markers.

Giriş ve Amaç: Biz bu çalışmamızda, farklı Gleason skoruna sahip insan prostat kanseri dokularında Lipokalin-2 (NGAL-nötrofil jelatinaz ilişkili lipokalin) ekspresyon seviyelerini tespit etmek ve ekspresyon seviyeleri ile dosetaksel tedavisine yanıt, Gleason skoru, progresyonsuz sağkalım ve genel sağkalımı içeren klinikopatolojik parametreler arasında bir ilişki olup olmadığını araştırmayı amaçladık. Materyal-Metod: Metastatik kastrasyon dirençli prostat kanseri için en az üç kür doketaksel alan ve klinik sonuçları takip edilen 33 hasta çalışmaya dahil edildi. Hastaların parafin blokları retrospektif olarak Çanakkale Onsekiz Mart Üniversitesi Tıp Fakültesi Hastanesi ve Çanakkale Mehmet Akif Ersoy Devlet Hastanesi patoloji arşivlerinden elde edildi. Lipokalin-2 ekspresyon durumu, anti-Lipokalin-2 antikoru kullanılarak immünohistokimyasal (IHC) yöntem ile belirlendi. Bulgular: Çalışma grubunda Lipokalin-2 yaklaşık %37 oranında eksprese edilmiştir. Lipokalin-2 ekspresyonu pozitifliği, yüksek Gleason skoru ile anlamlı derecede ilişkiliydi (P = 0.027). Lipokalin-2 ekspresyonu ile dosetaksel yanıtı arasında anlamlı bir ilişki yoktu. Lipokalin-2 ekspresyonu pozitif olan grupta hastaların genel sağkalım ve progresyonsuz sağkalım süreleri numerik olarak daha kısa olmakla birlikte istatistiksel olarak anlamlılık saptanmadı. Bu klinikopatolojik çalışma, farklı kanser tiplerinde görülen yüksek lipokalin-2 ekspresyonunun prognoza olan olumsuz etkisini destekler niteliktedir.Sonuç: Verilerimiz, yüksek Lipokalin-2 ekspresyonunun yüksek Gleason skoru ve kısa sağkalım ile ilişkisi nedeniyle prostat kanserinde kötü prognoz ile ilişkili olabileceğini göstermektedir. Sonuç olarak bu çalışma Lipokalin-2 ekspresyonunun prostat kanserinde prediktif belirteçten ziyade ümit verici bir prognostik belirteç olabileceğini desteklemektedir.Anahtar Kelimeler: Prostat adenokarsinomu, Dosetaksel, Lipokalin-2, Gleason skoru, Prediktif ve Prognostik Belirteçler.
