Thesis, Van, 2018Stress is defined as the physical and psychological response that the body has given to adapt to changing situations; reveal behavioral responses at different scales. In this study, the effects of water extracts of Gundelia tournefortii L. (Kenger) plants on anxiety and motor activity were investigated in rats exposed to chronic immobilization stress. In the study, 5 groups were formed as 8 rats in each group, and the duration of the study was planned to be 30 days in total. In the study, group 1 was chosen as control group. The rats in group 2 were exposed to immobilization stress for 30 days 30 minutes after orally saline application. Gundelia tournefortii L. water extract (***mg/kg /day/po) was administered to rats in group 3 for 30 days. The rats in group 4 were exposed to immobilization stress for 30 days 30 minutes after administration of Gundelia tournefortii L. water extract (***mg/kg /day/po). The rats in group 5 were exposed to immobilization stress for 30 days after 30 minutes of application of Fluoxetine (10 mg/kg/day/po). At the end of the 30th day, all groups were administered a Rotarod (motor activity test) and elevated Plus Maze Test (anxiety test). After all tests were completed, routine biochemical tests (ALT, AST, LDH, Glucose, Total Protein, Uric acid, cholesterol, bilirubin) were performed on blood samples taken from rats under anesthesia. According to the results of the anxiety tests at the end of the 15th and 30th days of study, it was found that anxiolytic effects of Gundelia tournefortii L. plant was significant in the chronic stressed rats and Gundelia tournefortii L. plant showed better anxiolytic effects when compared to Fluoxetine used clinically for anxiolytic purposes. Rotarod test results showed that chronic stress or Gundelia tournefortii L. plant did not affect motor activity. Results of serum biochemical analyzes showed that Gundelia tournefortii L. plant did not affect blood biochemistry when administered alone or in combination with chronic immobilization stress; However, it was observed that Fluoxetine, used clinically for anxiolytic purposes, elevated liver enzymes and decreased total protein ratio.As a result, it has been concluded that Gundelia tournefortii L. plant extract may show a therapeutic effect on stress and complications due to stress.According to these results, the mechanism of action underlying the anxiolytic effects of Gundelia tournefortii L. plant is planned to be the target of our new studies in future periods.

(Kenger) Bitkisi Su Ekstraktının Etkisi, Yüksek Lisans Tezi, Van, 2018.Stres, bedenin değişen durumlara adaptasyon sağlamak için vermiş olduğu fiziksel ve psikolojik yanıt olarak tanımlanmakta olup; farklı derecelerde davranışsal cevapları ortaya çıkarmaktadır. Bu çalışmada kronik immobilizasyon stresine maruz bırakılan ratlarda anksiyete ve motor aktivite üzerine Gundelia tournefortii L. (Kenger) bitkisi su ekstraktının etkisi incelendi. Çalışmada her grupta 8 rat olacak şekilde 5 grup oluşturuldu ve çalışmanın süresi toplam 30 gün olarak planlandı. Çalışmada 1. grup kontrol grubu olarak seçildi. 2. gruptaki Ratlar oral yolla serum fizyolojik uygulanmasından 30 dakika sonra immobilizasyon stresine 30 gün süresince maruz bırakıldı. 3. gruptaki Ratlara Gundelia tournefortii L. su ekstraktı (***mg/kg/gün/po) 30 gün süresince uygulandı. 4. gruptaki Ratlara Gundelia tournefortii L. su ekstraktı (***mg/kg/gün/po) uygulanmasından 30 dakika sonra immobilizasyon stresine 30 gün süresince maruz bırakıldı. 5. gruptaki Ratlar ise Fluoksetin (10 mg/kg/gün/po) uygulanmasından 30 dakika sonra immobilizasyon stresine 30 gün boyunca maruz bırakıldılar. 30. gün sonunda tüm gruplara Rotarod (motor aktivite testi) ve Yükseltilmiş Artı Labirent Testleri (anksiyete testi) uygulandı. Tüm testler tamamlandıktan sonra Ratlardan anestezi altında alınan kan numunelerinde rutin biyokimya testleri (ALT, AST, LDH, Glukoz, Total Protein, Ürik Asit, Kolesterol, Bilurubin) yapıldı. Çalışmada gerek 15. gün, gerekse 30. günün sonunda yapılan anksiyete testleri sonuçlarına göre, kronik strese maruz kalan ratlarda, Gundelia tournefortii L. bitkisinin anksiyolitik etkisinin anlamlı olduğu, klinikte anksiyolitik amaçla kullanılan Fluoksetin ile kıyaslandığında Gundelia tournefortii L. bitkisinin daha iyi anksiyolitik etki gösterdiği belirlendi. Rotarod testi sonuçları kronik stresin veya Gundelia tournefortii L. bitkisinin motor aktiviteyi etkilemediğini gösterdi. Serum biyokimyasal analizlerin sonuçları ise, Gundelia tournefortii L. bitkisinin yalnız veya kronik immobilizasyon stresi ile birlikte uygulandığında kan biyokimyasını etkilemediği; ancak, anksiyolitik amaçla klinik kullanımda yer alan fluoksetinin karaciğer enzimlerini yükselttiği ve total protein oranını düşürdüğü gözlendi.Sonuç olarak Gundelia tournefortii L. bitki ekstresinin stres ve strese bağlı olarak gelişen komplikasyonlar üzerine iyileştirici etki gösterebileceği sonucuna varılmıştır.Bu sonuçlara göre Gundelia tournefortii L. bitkisinin anksiyolitik etkilerinin altında yatan etki mekanizmalarının aydınlatılması, ileriki dönemlerde yeni çalışmalarımızın hedefi olarak planlanmaktadır.
