In this study, the effects of Cynara scolymus L. leaf extract on oxidative stress and liver damage were investigated.Totally, there were 42, 4-month old Wistar male albino rats in 6 groups in the study design. Three groups did not receive CCl4; which were control group, olive oil control group receiving olive oil intraperitoneally and Cynara scolymus L. control group receiving artichoke leaf extract by gavage technique. Three other groups were intraperitoneally given CCl4 (0,2 ml/kg) dissolved in olive oil (1:1) for the first 10 days of the study, which were CCl4, CCl4 natural recovery and curative groups. Then, the following 14 days the curative group intragastrically received the Cynara scolymus L. (1,5 g/kg) leaf extract dissolved in distilled water by gavage technique and meanwhile CCl4 natural recovery group was left to recover itself. Afterwards, heart blood samples and liver tissue samples were collected from each rat in all the groups under anesthesia following the ethics regulation. The sample collection was carried out at the end of day 10 for CCl4 group and at the end of day 24 for the other groups. Moreover, aspartate aminotransferase (AST), alanin amino transferase (ALT), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) parameters were measured from blood serum samples; catalase and superoxide dismutase (SOD) activity and malondialdehyde (MDA) levels were measured from haemolysate and homogenate samples; and DNA fragmentation was analyzed from liver tissue samples.The test results showed that, after the liver damage, the curative group receiving the Cynara scolymus L. leaf extract had AST enzyme activity levels close to the control values, and ALT and ALP enzyme activity levels lower than the CCl4 group.The SOD inhibition and catalase activity measures of the haemolysate samples of the curative group were close to the control values. Also, the curative group MDA levels were lower than that of the CCl4 natural recovery group.The liver homogenate of the CCl4 group had decreased SOD inhibition measures. Curative group liver homogenate had decreased SOD inhibition measures to a lesser degree. In addition, the curative group liver homogenate MDA level was lower than that of the CCl4 natural recovery group. However, there was an increase in the catalase activity of the curative group liver homogenate.The results indicated that the DNA fragmentation was increased by CCl4 application and the fragmentation was decreased in the curative group receiving the Cynara scolymus L. leaf extract.Histological analysis provided evidence for Cynara scolymus L. leaf extract to decrease the liver damage induced by CCl4 application.Overall, the study showed that Cynara scolymus L. leaf extract has an important effect on decreasing the oxidative stress and liver damage induced by CCl4.

Bu araştırmada, sıçanlarda karbon tetrakloridin neden olduğu oksidatif stres ve karaciğer hasarı üzerine Cynara scolymus L. yaprak ekstraktının etkileri araştırıldı.Çalışmada 42 adet 4 aylık Wistar cinsi erkek albino sıçan kullanılarak 6 grup oluşturuldu. Kontrol grubu, intraperitonel yolla zeytinyağı verilen zeytinyağı kontrol grubu ve gavaj tekniği ile enginar yaprağı ekstraktı verilen Cynara scolymus L. kontrol grubu CCl4 uygulaması almadı. CCl4, CCl4 doğal iyileşme ve küratif grup deney hayvanlarına çalışmanın ilk 10 gününde zeytin yağında 1:1 oranında çözdürülen CCl4 (0,2 ml/kg) intraperitonel yolla verildi. CCl4 uygulaması sonrasında küratif gruptaki deney hayvanlarına 14 gün boyunca intragastrik olarak gavaj yöntemi ile Cynara scolymus L. yaprak ekstraktı (1,5 g/kg) distile suda çözündürülerek verildi. CCl4 grubu sıçanlardan 10. gün, diğer deney gruplarımızdaki sıçanlardan ise çalışmanın 24. günü tamamlandıktan sonra, etik kurallara uygun olarak anestezi altında, uygun teknikler kullanılarak kalp kanı ve karaciğer doku örnekleri alındı. Kan serumlarında aspartat aminotranferaz (AST), alanin amino transferaz (ALT), alkalen fosfataz (ALP) ve laktat dehidrogenaz (LDH); hemolizat ve homojenatta katalaz ve süperoksit dismutaz (SOD) aktivitesi ile malondialdehit (MDA) düzeyi ölçüldü. Karaciğer dokusunda DNA fragmantasyonu ölçümü yapıldı.Karaciğer hasarı oluşumu sonrasında Cynara scolymus L. yaprak ekstraktı verilen küratif grupta AST enzim aktivitesinin çok önemli düzeyde kontrole yaklaştığı, ALT ve ALP enzim aktivitelerinin ise CCl4 grubuna göre düşük olduğu belirlendi.Hemolizatta küratif grup SOD inhibisyonu ve katalaz aktivitesinin kontrole yaklaştığı; CCl4 doğal iyileşme grubuna göre küratif grup hemolizat MDA düzeyinin azaldığı belirlendi.Karaciğer homojenatında CCl4 uygulanan gruplarda SOD inhibisyonunda azalma bulundu. Küratif grupta SOD inhibisyonunda azalmanın daha az olduğu belirlendi. Küratif grup homojenat katalaz aktivitesinde artış belirlendi. Küratif grup homojenat MDA düzeyinin CCl4 doğal iyileşme grubuna göre düşük olduğu belirlendi.CCl4 kullanımı sonucunda DNA fragmantasyonunun arttığı ve Cynara scolymus L. yaprak ekstraktının kullanıldığı küratif grupta DNA fragmantasyonunun azaldığı belirlendi.Histolojik yönden incelendiğinde; CCl4 ile oluşturulan karaciğer hasarının Cynara scolymus yaprak ekstraktı kullanımı sonucunda azaldığı belirlendi.Sonuç olarak, karaciğerde hasara sebep olan karbon tetrakloridin neden olduğu oksidatif stres ve karaciğer hasarı üzerine Cynara scolymus L. yaprak ekstraktının önemli ölçüde hasarı azaltma yönünde etkili olduğu belirlendi.
