The prognosis of Malignant Pleural Mesothelioma (MPM) is generally poor, with a median survival time of around 12 months. The purpose of this study is to examine the gene expression profiles and the usability of the gene expression profiles as prognostic biomarkers of midkine (MKN), syndecan-1 (SOC-1), hyaluronan synthase-2 (HAS-2), fibulin-3 selected from the literature and sestrin-1 (SESN-1) and laminin subunit alpha-4 (LAMA-4) determined with Unsupervised Survival Analysis Tool (USAT) and two housekeeping genes, glucose-6-phosphate dehydrogenase (G6PD), TATA-box transporter protein (TBP), which were determined by means of qPCR method in 54 patients with definite diagnosis and chemotherapy treatment. The relationship between gene expression and prognosis was assessed by categorizing the samples into two groups using all probability cut-off values using the LMRC method. p values were below 0.05 and the predictive value was considered significant between 10th and 90th percentile. It was determined that overexpression of MKN (p=0.002), HAS-2 (p=0.006) and SESN-1 (p=0.005) in MPM patients was associated with survival time in multivariate analysis. In addition, it was determined that HAS-2 (p=0.014) and SESN-1 (p=0.045) overexpression of genes in epithelial type MPM patients were associated with survival in multivariate analysis. In conclusion, in patients with MPM, MKN over-expression was associated with poor prognosis and HAS-2 and SESN-1 over-expression were associated with good prognosis. It has also been suggested that HAS-2 and SESN-1 over-gene expressions may be useful as biomarkers for a good prognosis in patients with MPM with epithelial cell type. It will also contribute to our knowledge about the evaluation of such markers in other biological enviroments in larger groups, and to develop more accurate convictions for the same purposes.

Malign Plevral Mezotelyoma (MPM) 'nin prognozu iyi değildir, ortanca yaşam süresi 12 ay civarında verilmektedir. Bu çalışmada MPM kesin teşhisi konmuş ve kemoterapi tedavisi alan 54 hastanın tümör dokusunda Real Time Polymerase Chain Reaction (qPCR) yöntemiyle, midkine (MKN), syndecan-1 (SOC-1) ve hyaluronan synthase-2 (HAS-2); literatürden seçilen fibulin-3; ayrıca Unsupervised Survival Analysis Tool (USAT) ile belirlenen sestrin-1 (SESN-1) ile laminin subunit alpha-4 (LAMA-4) ve iki housekeeping gen; glukoz-6-fosfat dehidrogenaz (G6PD), TATA-box taşıyıcı protein (TBP)'nin gen ifadelerinin incelenmesi ve saptanacak gen ifade profillerinin prognoz biyobelirteci olarak kullanılabilirliklerinin araştırılması amaçlanmıştır. Gen ifadesi ve prognoz arasındaki ilişki Log Rank multiple cut-off analysis (LMRC) yöntemiyle tüm olasılıklı kestirim (cut-off) değerleri kullanılarak, örnekler iki gruba katogorize edilerek değerlendirildi. p değerleri 0.05'in altında ve kestirim değeri 10-90 percantil arası anlamlı kabul edildi. MPM hastalarında MKN (p=0.002), HAS-2 (p=0.006), SESN-1 (p=0.005) aşırı gen ifadelerinin çoklu değişken analizlerde sağkalım zamanı ile ilişkili olduğu belirlendi. Buna ek olarak, epiteloid tip MPM'li hastalarda HAS-2 (p=0.014) ve SESN-1 (p=0.045) aşırı gen ifadelerinin çoklu değişken analizlerde sağkalım ile ilişkili olduğu belirlendi.
