This study was conducted to determine the effects of the arsenic, lead, and cadmium mixtures on `Saban`, a variety of bread wheat, and `Hasat`, a variety of barley, which were registered by Trakya Agricultural Research Institute in 2014.Barley and wheat seeds were grown in the plant growth cabinet at 20ºC for 10 days by applying photoperiod. At the end of the 10th day, while the control group was watered with distilled water, among the other groups, the 1st day group and the 5th day group were watered with the freshly prepared solutions of the mixture containing arsenic, lead, and cadmium at the concentrations of 15 μM, 30 μM, and 60 μM for 1 day and 5 days, respectively. The germination percentages, root stem lengths, root stem wet and dry weights, total protein contents, some hormone levels, and MDA contents in barley and wheat plants were measured. How the expression levels of SOD, CAT, GS, and GPX antioxidant enzymes in plants are affected during metal stress has been investigated. The germination of plants for the study was performed at T.U. Faculty of Science, Department of Biology. Heavy metal and enzyme expression analyses in plants were carried out at Trakya University Research and Development Center (TUTAGEM).A decrease was observed in the experimental groups, in which 15 μM, 30 μM, and 60 μM (arsenic, lead, and cadmium) metal ion mixture was applied, in terms of the germination rate depending on the increased concentration, and a decrease was observed in the root and stem dry weights of the plants in all groups compared to the control group. MDA levels were determined to increase at all doses. It was determined that heavy metals accumulated by increasing in the tissues due to the increased concentration of heavy metals in the heavy metal ion-applied groups compared to the control group. As a result, it was determined in the study that there were significant changes in the expression levels of antioxidant enzymes, which are the primary response to oxidative stress in plants exposed to heavy metals, depending on the stress.

Araştırma Trakya Tarımsal Araştırma Enstitüsü tarafından 2014 yılında tescil edilen bir ekmeklik buğday çeşidi olan `Saban` ve bir arpa çeşidi olan `Hasat` üzerinde arsenik, kurşun ve kadmiyum karışımlarının etkisini belirlemek amacıyla yapılmıştır.Arpa ve buğday tohumları 20 ºC'de fotoperiyot uygulanarak bitki büyüme kabininde 10 gün süreyle büyütüldü.10. günün sonunda, kontrol grubu distile su ile sulanırken, diğer gruplar 1. gün grubu 1 gün süre ile 5. gün grubu 5 gün süre ile taze hazırlanmış 15 μM, 30 μM ve 60 μM konsantrasyonlar da arsenik, kurşun ve kadmiyum içeren karışım solüsyonları ile sulandı. Arpa ve buğday bitkilerinde çimlenme yüzdeleri, kök gövde uzunlukları, kök gövde taze ve kuru ağırlıkları, total protein miktarları, bazı hormon seviyeleri ve MDA içerikleri ölçüldü. Bitkilerdeki SOD, CAT, GS ve GPX antioksidan enzimlerinin metal stresi sırasında ekspresyon seviyelerinin nasıl etkilendiği araştırıldı. Araştırma için bitkilerin çimlendirilmesi T.Ü Fen Fakültesi Biyoloji Bölümünde yapıldı. Bitkilerdeki ağır metal ve enzim ekspresyon analizleri Trakya Üniversitesi Araştırma ve Geliştirme Merkezi (TUTAGEM)'inde gerçekleştirildi.15 μM, 30 μM ve 60 μM (Arsenik, kurşun ve kadmiyum) metal iyonu karışımı uygulanan deney gruplarında konsantrasyon artışına bağlı olarak çimlenme oranında, bitkilerin kök ve gövde kuru ağırlıklarında tüm gruplarda kontrol grubuna göre azalma gözlendi. MDA seviyelerinin tüm dozlarda yükseldiği tespit edildi. Ağır metal iyonu uygulanan gruplarda kontrol grubuna göre ağır metallerin konsantrasyon artışına bağlı olarak dokularda artarak biriktiği tespit edildi. Sonuç olarak çalışmada ağır metale maruz kalan bitkide oksidan stresin primer cevabı olan antioksidan enzimlerin ekspresyon seviyelerinde strese bağlı olarak anlamlı değişikliklerin olduğu belirlendi.
