The free radicals that present in the human organism damages the molecules like lipids, proteins and nucleic acids. On the other hand, antioxidant compounds have the ability to neutralize these free radicals and also they can have a major role in prevention of cancer, cataract, heart and brain disease and rheumatism. Thus, in our study, it is tried to identify the antioxidant effect of the Vanillic acid, which is the oxidized form of vanillin, against the oxidative effect of a well known alkylating chemical and antineoplastic agent mitomycin C (MMC). For this purpose, human blood lymphocyte cultures that are taken from healthy donors are used. In vitro micronucleus (MN) and COMET tests are used in order to determine the oxidative damage and antioxidant effect in the level of DNA and chromosome. In this study, 0,25 µg/ml mitomycin and 1 and 2 µg/ml dosage of vanillic acid was used. The DMSO was used in the solution of vanillic acid and mitomycin C was used as a negative control. 2 µg/ml dosage of vanillic acid formed substantive MN and damaged DNA in the micronucleus and COMET tests. Mitomycin C has significantly increased the rate of damaged DNA and cells with micronucleus. On the other hand 1 µg/ml dosage of vanillic acid has significantly decreased the rate of DNA damaged cells and the frequency of micronucleated cells. In fact, this dosage of vanillic acid has reduced the rate of cells with micronucleus that are formed with mitomycin C and the genetic damage index rate in COMET assay to DMSO control level. However because of 2 µg/ml vanillic acid itself increased the rate of micronucleus and DNA damage, it couldn?t decrease the damage that is caused by mitomycin C. In conclusion, it is proved that vanillic acid which is an vegetative phenolic compund could prevent the oxidative DNA damage and damages in chromosomes when used in proper low dosages.

İnsan organizmasında bulunan serbest radikaller; yağlar, proteinler ve nükleik asitler gibi farklı moleküllerin oksidatif olarak bozulmasına neden olmaktadır. Antioksidan bileşikler ise bu serbest radikalleri etkisiz hale getirme yeteneğine sahiptir ve aynı zamanda; kanser, katarakt, kalp damar hastalıkları, beyin rahatsızlıkları ve romatizmanın önlenmesinde temel bir rol oynayabilirler. Buradan yola çıkarak çalışmamızda hücrede oksidatif hasar oluştuğu iyi bilinen alkilleyici bir kimyasal ajan olan ve antineoplastik ajan olarak da kullanım alanına sahip mitomisin C (MMC) `nin oksidatif etkisine karşı vanillinin okside formu vanillik asidin antioksidan etkisi belirlenmeye çalışılmıştır. Bu amaçla sağlıklı dönorlardan alınan insan kan lenfosit kültürleri kullanılmıştır. Deoksribonükleik asit (DNA) ve kromozom düzeyindeki oksidatif hasarı ve antioksidan etkiyi belirlemek amacıyla in vitro Mikronükleus (MN) ve COMET testleri gerçekleştirilmiştir. Çalışmada mitomisin'in 0,25 µg/ml dozu, vanillik asidin ise 1 ve 2 µg/ml' lik dozları kullanılmıştır. Vanillik asit ve mitomisin C' nin çözülmesinde kullanılan Dimetilsülfoksit (DMSO) negatif kontrol olarak kullanılmıştır. Vanillik asidin 2 µg/ml dozu Mikronükleus ve COMET testlerinde anlamlı MN ve hasarlı DNA oluşturmuştur. Mitomisin C' de tek başına mikronükleuslu hücre ve hasarlı DNA oranını anlamlı arttırmıştır. 1 µg/ml'lik vanillik asit ise her iki test sisteminde mitomisin C'nin oluşturduğu mikronükleuslu DNA hasarlı hücre oranını anlamlı azaltmıştır. Hatta vanillik asidin bu dozu mitomisin C ile oluşturulan mikronükleuslu hücre ve COMET testindeki genetik hasar indeksi oranını DMSO kontrol düzeyine indirmiştir. 2 µg/ml vanillik ise kendi başına da mikronükleus ve DNA hasar oranını arttırdığı için mitomisin C' nin oluşturduğu hasarı yeterli seviyede azaltamamıştır. Sonuç olarak antioksidan bir bitkisel fenolik bileşik olan vanillik asidin uygun düşük dozlarda kullanıldığında oksidatif DNA hasarı ve kromozomlardaki hasarları engelleyebileceği öngörülmüştür.
