The effect of nigella sativa oil on serum brain derived neurotrophic factor (BDNF), biogenic amines and oxidative stress in the rat induced metabolic syndromeIt is considered that all over the world, especially in the developed countries, the metabolic syndrome, growing with every passing day, the most important factor in the pathogenesis of which is known to be the insulin resistance, arises in connection with the effects of genetic and environmental factors. One of the reasons of the growing of the prevalence of the metabolic syndrome is known to be the fructose, taken at a high level within a diet which is a separate risk determinant. In this study, we had the purpose to contribute to the literature with the data to be obtained from investigating the mechanism of the reason of increase of the development and the prevalence of the Nigella sativa oil metabolic syndrome, BDNF, biogenic amines and oxidative stress levels, having a metabolic syndrome formed in rats with a fructose diet. In the study, 21 male Sprague-Dawley rats about weight of 200-240 g have been used. The rats were seperated to 3 groups, each of which has 7 rats. Group1; control group (10 weeks), group 2; metabolic syndrome with fructose (10 weeks), group3; given Nigella sativa oil after metabolic syndrome progress (10+4 week) in created. Rats were decapited after the study. Blood samples were taken, the serum glucose, insulin, total cholesterol, triglycerid, HDL, LDL, VLDL levels, TAS and TOS, biogenic amines (dopamine, noradrenaline, adrenaline, serotonin, 5- HIAA), cytokines (IL-6, TNF-α), BDNF levels were measured in the serum and the effects of Nigella sativa oil on these parameters were examined. Serum insulin, glucose, HOMA-IR, triglyceride, total cholesterol, LDL-C, TNF-α, TOS, dopamine and noradrenaline levels measuring were compared to the control group found statistically significantly higher and the HDL-C, serotonin amount was compared to the control group found significantly lower in the metabolic syndrome groups (P<0,05). After the formation of metabolic syndrome, that we gave the Nigella sativa oil group serum glukoz, insülin, HOMA-IR and lipid profile compared to the control group levels significantly different were lower (p<0,05). Formation of metabolic syndrome, that we gave the Nigella sativa oil group TOS, TNF-α ve IL-6 levels were lower, but the decrease did not have a statistical significance (p>0,05). Metabolic syndrome group BDNF levels were compared to the control group lower, but the decrease did not have a statistical significance (p>0,05). Formation of metabolic syndrome, that we gave the Nigella sativa oil group BDNF levels were compared to the metabolic syndrome group found statistically significantly higher.According to data obtained as a result of the study, Mann Whitney-U test and Kruskal Wallis the value of p<0.05 was used as a level of statistical significance in pair wise comparisons between the groups.Consequently, Nigella sativa oil have a positive effect the serum BDNF, biogenic amines, total oxidative status, total antioxidant status levels, cytokines, insulin metabolism and lipid profile which can be useful to in the patients with metabolic syndrome and it looks like a promising option has been concluded.Key words: Metabolic syndrome, Nigella sativa oil, Brain derived neurotrophic factor (BDNF).

Gelişmiş ülkeler başta olmak üzere bütün dünyada her geçen gün artan, patogenezinde en önemli faktörün insülin rezistansı olduğu bilinen metabolik sendromun, genetik ve çevresel faktörlerin etkisi ile bağlantılı olarak ortaya çıktığı düşünülmektedir. Metabolik sendrom prevalansının artış nedenlerinden biri de ayrı bir risk belirleyicisi olan yüksek düzeyde diyet içinde alınan fruktoz olduğu bilinmektedir. Biz bu çalışmada; fruktoz diyeti ile metabolik sendrom oluşturulmuş sıçanlarda, Nigella sativa yağı'nın metabolik sendromun ve prevalansının artış nedenlerindeki mekanizmalara etkilerini araştırıp, BDNF, biyojenik aminler ve oksidatif stres düzeylerinin elde edilecek veriler ile literatüre katkıda bulunmayı amaçladık.Çalışmada 200-240 gr arasında 21 adet erkek Sprague-Dawley cinsi sıçan kullanıldı. Sıçanlar her bir grupta 7 sıçan olacak şekilde 3 gruba ayrıldı. Grup 1; kontrol gurubu (10 hafta), grup 2; fruktoz ile metabolik sendrom oluşturulan grup (10 hafta), grup 3; metabolik sendrom oluştuktan sonra Nigella sativa yağı verilen grup (10+4 hafta) olarak düzenlendi. Çalışma tamamlandıktan sonra sıçanlar dekapite edilerek alınan kan örneklerinden; serum glukoz, insülin, total kolesterol, trigliserid, HDL, LDL, VLDL düzeyleri, TAS ve TOS, biyojenik aminler (dopamin, noradrenalin, adrenalin, serotonin, 5- HIAA), sitokinler (IL-6, TNF-α), BDNF düzeyleri ölçüldü ve Nigella sativa yağının bu parametreler üzerine etkileri incelendi. Metabolik sendrom grubunda serum insülin, glukoz, HOMA-IR, trigliserid, total kolesterol, LDL-K, TNF-α, TOS, noradrenalin, dopamin düzeyleri ölçüldüğünde, kontrol grubuna göre istatistiksel olarak anlamlı yüksek iken; HDL-K ve serotonin miktarlarının kontrole göre anlamlı şekilde azaldığı tespit edilmiştir (P<0,05). Serum glukoz, insülin, HOMA-IR ve lipid profili düzeylerinin, metabolik sendrom oluştuktan sonra Nigella sativa yağı verdiğimiz grupta metabolik sendrom grubuna göre anlamlı olarak azaldığı tespit edildi (p<0.05). Metabolik sendrom oluştuktan sonra Nigella sativa yağı verdiğimiz grupta TOS, TNF-α ve IL-6 değerlerinde metabolik sendrom grubuna kıyasla düşüş gözlendi, fakat bu düşüş istatistiksel olarak anlamlı değildi (P>0.05). Metabolik sendrom grubunda, BDNF düzeylerinde kontrole kıyasla düşüş gözlendi, fakat bu düşüş istatistiksel olarak anlamlı değildi (P>0.05).. Metabolik sendrom oluştuktan sonra Nigella sativa yağı verdiğimiz grupta BDNF düzeyleri ise metabolik sendrom grubuna göre istatistiksel olarak anlamlı şekilde yüksek bulundu (p<0,05).Çalışma sonucu elde edilen verilere göre gruplar arasında Kruskal Wallis, ikili karşılaştırmalarda Mann Whitney-U testi ve en düşük istatistiksel anlamlılık düzeyi olarak p<0.05 değeri kullanıldı.Sonuç olarak, Nigella sativa yağının serum BDNF, biyojenik aminler, total oksidatif durum, total antioksidan durum düzeyleri, sitokinler, insülin metabolizması ve lipit profili üzerine olumlu etki yapabildiği ve metabolic sendromlu hastalarda kullanımının yararlı olabileceği kanısına varılmıştır.Anahtar Kelimeler: Metabolik sendrom, Nigella sativa yağı, beyin kaynaklı nörotrofik faktör (BDNF).
