Abstract

Multiple sclerosis (MS) is one of the central nervous system disorder and the most common inflammatory disease. MS's etiology is unknown. Most probably MS is triggered by environmental factors in genetically susceptible persons. Genes and environmental' roles in MS are not understood clearly. Lots of GWAS (Genome Wide Association Study) studies are done for genes related with MS. According to these studies genetic variations in MS mostly found in HLA (Human Leukocyte Antigen) genes part of MHC (Major Histocompatibility Complex) region of chromosome 6p21.3. Moreover, MS related genes also found in non- HLA regions. These genes are IL2RA, IL7R, CLEC16A, CD58, CD226, KIF1B, etc. IL2RA (interleukin Receptor 2 alpha) has highest relation with MS is the class of non- HLA MS related genes. There are only 2 variations in OMIM related with MS, and both of them are IL2RA variations. Therefore, IL2RA has a key role in MS. In this thesis we aimed to sequence of complete IL2RA gene in the 5 Turkish patients and to find known and unknown variations with the help of Next Generation Sequencing (NGS). To reach target we sequenced 5 patients 80 KB IL2RA gene two times and we listed all variations found. All variations found were annotated and all de novo variations determined according to dbSNP (database SNP). In results, we detect lots of de novo indel and SNPs including putative frameshift mutation in mRNA coding region. We will confirm all these variations by using Sanger sequencing.

Multipl Skleroz (MS) en yaygın merkezi sinir sistemi inflamatuar hastalığıdır. Multipl skleroz 'un etiyolojisi bilinmemektedir. Büyük ihtimalle MS genetik yatkınlığı olan bireylerde çevresel faktörlerin tetiklemesi ile ortaya çıkar. MS'deki genlerin ve çevrenin etkisi tam olarak anlaşılamamıştır. MS'de etkili olan genler için pek çok GWAS (Genom Wide Association Study) yapılmıştır. Bu çalışmalara göre MS'den ilişkilendirilmiş varyasyonlar daha çok kromozom 6p21.3 deki MHC (Major Histocompatibility Complex) bölgelerinde tespit edilmişti ve bunlar HLA(Human Leukocyte Antigen) genleridir. Ayrıca MS HLA olmayan bölgelerde MS'le ilişkili genler keşfedilmiştir. Bunlar IL2RA, IL7R, CLEC16A, CD58, CD226, KIF1B, vb. genlerdir. IL2RA (interleukin Receptor 2 alpha) geni MS'le ilişkilendirilen ve HLA bölgesinde olmayan genlerden istatistiksel olarak en yüksek yatkınlık değerine sahip olan gendir. OMIM (Online Mendelian Inheritance in Man) veri tabanında MS'le ilişkilendirilmiş varyasyonlar sadece IL2RA geninde bulunmaktadır. Bu açıdan IL2RA MS'de önemli bir yere sahiptir. Bu tezde IL2RA geninin 5 adet Türk MS hastalarında Yeni Nesil Dizileme(YND) yöntemleri kullanılarak tüm gen dizilemesi yapılması ve bilinen ve bilinmeyen varyasyonların tespit edilmesi amaçlanmıştır. Hedef kapsamında gerçekleştirilen çalışmadan 5 adet hastanın 80 kb (kilo baz)'lik IL2RA geni ikişer kere dizilenmiş ve bütün varyasyonlar listelenmiştir. Bulunan bütün varyasyonlar dbSNP (database SNP)'e göre annote edilmiş ve dbSNP 'de olmayanlar aday varyasyon olarak belirlenmiştir. Sonuçlara göre mRNA bölgesinde frameshift mutasyona sebep olan yeni bir adayı varyasyon dahil olmak üzere pek çok SNP, indel ve de novo varyasyon tespit edilmiştir.
