Heat stress is often defined as the rise in temperature beyond a threshold level for a period of time sufficient to cause irreversible damage to plant growth and development. From this side of view, heat tolerance is generally defined as the ability of the plant to grow and produce economic yield under high temperatures. The present study was performed in the laboratories of faculty of Science, Ain Shams University and Field Crops Research Institute (FCRI), Agricultural Research Center (ARC), Giza, Egypt. The present study aimed to estimate the differences among maize hybrids in relation to heat stress and find out some biochemical markers for heat tolerance in Egyptian maize hybrids. Seeds of the sixteen Egyptian maize hybrids were germinated for 10 days under normal conditions, a set of 16 hybrid seedlings was subjected to 40oC, leaf samples were obtained after 24, 48 and 72 hours after treatment initiation. Shoot length, root length, plant height, seedling fresh weight and survival percentage were recorded. Two maize hybrids were selected as tolerant and two as sensitive, seedlings of the four hybrids were subjected to heat shock at 43 and 47oC for two hours and leaf samples were obtained. Enzyme activity and protein content were quantified in addition to SDS-PAGE protein electrophoresis. Oxidative enzymes activity was found to increase as heat temperature increase. Three bands of molecular weight of ~31.00, ~ 18.00 and ~ 16.00 kDa were induced under heat stress conditions. An induced band of ~ 100.00 kDa was induced under heat treatment in the two sensitive hybrids which could be used as negative marker for heat tolerance.

يعرف الضغط الحراري علي أنه الإرتفاع في درجات الحرارة إلي مستويات أعلي من المستوي الحدي الذي يتحمله النبات ولفترات كافيه لإحداث تأثير ضار وغير قابل للإرتداد علي نمو وتطور النبات. ومن هذا المنظور فإن تحمل الإجهاد الحراري يعرف عادة بمدي قدرة النبات علي النمو وإنتاج محصول إقتصادي تحت الحراره العالية. نفذت هذه التجربه بصوب ومعامل كل من كلية العلوم –جامعة عين شمس ومعهد بحوث المحاصيل الحقليه- مركز البحوث الزراعية بالجيزة, وذلك بهدف تقدير الفروق في قدره الهجن المصريه علي تحمل الإجهاد الحراري وإيجاد بعض الواسمات البيوكيميائيه لصفة التحمل فيها. تم إنبات 16 هجين من الّذرة الشاميه لمدة عشرة أيام تحت الظروف الطبيعية ثم تم تعريض مجموعات كل منها يضم الستة عشرهجينا تحت الإختبار لدرجة حراره 40 درجه مئويه لفترات 24,48,72 ساعه. تم تقدير طول الساق والجذر والوزن الغض للبادرات وكذلك نسبه اليادرات التي بقيت حيه بعد المعامله. بناء علي الاختلافات المعنوية بين الهجن تم اختيار هجينين اثنين كأكثر الهجن تحملا للحرارة وهجينين اثنين كأكثرها حساسيه حيث تم تعريضها لصدمه حراريه عند درجات حراره 43 , 47 درجة مئويه لمدة ساعتين. قدر النشاط الإنزيمي كميا لبعض الانزيمات وكذلك المحتوي الكلي من البروتينات إلي جانب التقدبر الكيفي للبروتين يطريقة التفريد الكهربي SDS-PAGE حيث أظهرت النتائج زياده في نشاط إنزيمات الاكسده المختبرة في الهجن المتحمله للحراره عنها في الهجن الحساسه. كما أظهر التفريد الكهربي للبروتين وجود ثلاث حزم ذات أوزان جزيئيه ّ31, 18, 16 كيلو دالتون والتي استحدثت كنتيجه للصدمه الحرارية بينما اختفت حزمه بروتينيه ذات وزن جزيئي 100 كيلو دالتون مفارنة بالعينه الضابطه.
