The current study aimed to extraction and purification of the protease inhibitor and its antimicrobial activity. The protease inhibitor was extracted from Lathyrus sativus and it's inhibited the trypsin enzyme with maximum percent of inhibition (89.6%), the crude extract prepared in 0.1M potassium phosphate buffer, pH 7 with showed maximum inhibitor activity (98%). The optimum extraction ratio efficiency was founded when the extraction ratio 1:7.5 (w:v) after 1h the maximum inhibitor activity obtained was (96%). The antimicrobial activity of protease inhibitor was estimated against seven bacterial & fungal strains, the maximum inhibition zone estimated 28 mm against Kluyveromyces marixians. Protease inhibitor, isolated from Lathyrus sativuswas purified by purification techniques, included ammonium sulphate percipitation 60%,followed by Ion exchange chromatography using DEAE–Cellulose column. The enzyme was purified to 2.5 times in the last step with an enzyme yield of 53.4%.

هدفت الدراسة الحالية إلى استخلاص مثبط إنزيم البروتييز وتنقيته ودراسة تأثيره ضد الميكروبي ، حيث استخلص مثبط إنزيم البروتييز من نبات الهرطمان الذي ثبط إنزيم التربسين بفعالية تثبيطية تقدر %89.6 ، استخلص المثبط باستخدام المحلول الدارئ لفوسفات البوتاسيوم بتركيز 0.1 مولر و(pH 7) ، حيث كانت الفعالية التثبيطية لإنزيم التربسين 98% ، قدرت نسبة المحلول الدارئ المثلى المستخدم لاستخلص المثبط بـ 1: 7.5 (و : ح) بعد ساعة واحدة من الاستخلاص ، حيث بلغت قيمة الفعالية التثبيطية للإنزيم 96% ، كما قدرت الفعالية الضد ميكروبية لمثبط البروتييز، إذا بلغ قطر منطقة التثبيط 28 ملم لخميرة Kluyveromyces marixians . تم تنقية المثبط البروتييزي بخطوتين: باستخدام التركيز بكبريتات الأمونيوم بنسبة إشباع 60% وكروماتوغرافيا التبادل الأيوني باستخدام عمود DEAE–Cellulose ، حيث بلغت عدد مرات التنقية للخطوة الأخيرة 2.5 بحصيلة إنزيمية بلغت 53.4 %.
